Backgrounds:As the most common primary intracranial tumors,glioma has the ability to diffusely invade the normal brain tissue.The patients have poor prognosis, especially for patients with glioblastoma, even if received the best treatments,the median survival is only 12 to 15 months. With regard to the diagnosis of glioma, imaging and pathological biopsy are the mainly methods,there is still no specific tumor marker.Osteopontin(OPN) is a phosphorylated glycoprotein,believed to be involved in a variety of physiological cellular functions.It has been shown to play an important role in tumorgenesis,tumor invasion,metastasis and angiogenisis in breast, lung, prostate,and colon cancers which were reported to have high expression of OPN. There are only limited data concerning the relationship between OPN and glioma.We investigate the possibility of OPN being a serum bio-marker for glioma by examining the serum OPN level in glioma patients.Objective:To examine the serum OPN level in glioma,meningiomas patients and healthy controls and to evaluate its clinical significance as a serum bio-marker for glioma.Methods:The serum OPN level was measured by enzyme linked immunosorbent assay(ELISA) in 49 cases of gliomas(grade II 20,gradeâ…¢14 and gradeâ…£15),16 cases of meningiomas and 20 cases of healthy controls.Results:Serum OPN level in glioma was significantly higher than that in meningiomas(P=0.001) and controls(P<0.001), the OPN level in meningiomas was significantly higher than that in controls(P<0.001); Serum OPN level in gradeâ…£of glioma was significantly higher than that in gradeIII(P<0.001), grade II (P<0.001), meningiomas(P<0.001) and controls(P<0.001), Serum OPN level in gradeâ…¢, grade II of glioma and meningiomas was significantly higher than that in controls(P< 0.001, P<0.001, P<0.001).There was no singificant difference between gradeâ…¢and grade II (P=0.970),grade II and meningiomas (P=0.999),gradeâ…¢and meningiomas (P=0.483).Conclusion:The serum OPN level of glioma and meningiomas was higher than healthy controls,it increased in a grade-dependent manner,may be a potential bio-marker for the diagnosis and prognosis of glioma.
