The relationship between GRP78 expression with clinicopathologic features and prognosis was analyzed.Results:The positive rate of GRP78 in esophagus squamous cell carcinomas tissues was significantly higher than that in normal esophageal squamous epithelium tissues (P<0.01). The positive rate of GRP78 in squamous cell carcinomas tissues of the esophagus was correlated to infiltration depth, cancer differentiation level and lymphatic metastasis (P<0.05), but not with gender and length of neoplasm (P> 0.05). A lower expreesion of GRP78 was observed in esophagus squamous cell carcinomas with longer-term survival (P<0.05).Conclusion:The positive expression of GRP78 were more common in patients with esophageal squamous cell carcinoma, which show that GRP78 may correlate to the occurrence and progress of esophagus squamous cell carcinomas. A higher expreesion of GRP78 in esophagus squamous cell carcinomas was one of the symbols of a poor prognosis.Part2:Expression of GRP78 mRNA and protein in esophageal squamous cell carcinoma.Objective:To investigate the expression of glucose-regulated proteins 78(GRP78) in human esophagus squamous cell carcinomas and the relationship between GRP78 and clinicopathologic features.Methods:fifty-nine cases of human esophagus squamous cell carcinoma specimens and twenty cases of adjacent normal tissues were collected from the patients who received operation for esophagus squamous cell carcinomas in Jinan Central Hospital Affiliated to Shandong University between 2007 and 2008. Semi-quantitative RT-PCR assay and Western-blotting were used to detect GRP78 mRNA and GRP78 protein expression respectively. The relationship between GRP78 expression with clinical parameters, such as gender,length of neoplasm,infiltration depth,cancer differentiation level,lymphatic-cmetastasis, was analyzed.Results:GRP78 mRNA and GRP78 protein were both expressed in human esophagus squamous cell carcinomas and normal esophagus tissues.There was a significant overexpression of GRP78 mRNA and protein in squamous cell carcinoma tissues of the esophagus as compared with normal esophagus tissues. The differences in expression of GRP78 between the two tissues were significant (P<0.01) Furthermore, the over-expression of GRP78 in squamous cell carcinoma tissues of the esophagus was correlated to infiltration depth, grade of differentiation and lymphatic metastasis (P<0.05), but not with sex and the length of neoplasm (P>0.05).Conclusion:GRP78 may correlate to the occurrence and progress of squamous cell carcinomas of the esophagus.
