Objective:Research on preparation of mouse models of hypertensive disorder complicating pregnancy(HDCP) through electrical stimulation.The localization and expression of transforming growth factor-β1(TGF-β1) and insulin-like growth factor-Ⅱ(IGF-Ⅱ) in the electrical stimulation-induced hypertensive disorder complicating pregnancy model mice placenta tissues are studied,and the relevance among TGF-β1, IGF-Ⅱand outbreak of hypertensive disorder complicating pregnancy is discussed.The influence of different treatment methods of L-arginine and magnesium sulfate on hypertensive disorder complicating pregnancy model mice is studied.Medthods:Mouse hypertensive disorder complicating pregnancy models were established with electrical stimulation induction,blood pressure,urine protein and other indicators of pregnant mice were tested,and appropriate hypertensive disorder complicating pregnancy model mice were selected. The model mice accepted different treatment methods of L-arginine and magnesium sulfate for intervention,the indicators such as blood pressure, urine protein level,plasma,NO concentration and nitric oxide synthase (NOS) activity in placenta,expression of TGF-β1 and IGF-Ⅱin placenta and the like of pregnant rats before and after intervention were observed, and the influence of different treatments on the indicators was compared.Results:1.Compared with the normal control group,blood pressure and urine protein levels of mice in the model group were increased(P<0.05, P<0.05),histopathological examination accorded with clinicopathological characteristics of hypertensive disorder complicating pregnancy.2.After administration treatment,the blood pressure and urine protein levels of pregnant mice in the L-arginine group,the magnesium sulfate group and the(L-arginine+sulfate) group were significantly lower than the model group(P<0.01,P<0.01),but were still higher than the control group(P<0.05,P<0.05).3.The NO concentration of plasma and placenta of pregnant mice in the L-arginine group and the(L-arginine+magnesium sulfate) group were significantly increased compared with the model group(P<0.05,P<0.01);Total NOS activity of placenta in the(L-arginine+magnesium sulfate) group was significantly increased compared with the model group or the magnesium sulfate group(P<0.05,P<0.05).4.TGF-β1 and IGF-Ⅱprotein were mainly expressed in endochylema of the placental villus trophoblastic cells and vascular endothelial cells,the expression level of TGF-β1 protein in(L-arginine+ magnesium sulfate) group was significantly lower than the model group (P<0.05),and IGF-Ⅱexpression was significantly higher than the model group(P<0.05).Conclusion:1.Hypertensive disorder complicating pregnancy mouse models can be established through electrical stimulation,NO level expression abnormity is an important factor of hypertensive disorder complicating pregnancy.2.TGF-β1 was over expressed and IGF-Ⅱwas inadequately expressed in placenta tissues,which is closely related with outbreak of hypertensive disorder complicating pregnancy,they may participate in the nosogenesis of hypertensive disorder complicating pregnancy and also may be results of pathophysiology changes of hypertensive disorder complicating pregnancy.3.Three treatments of L-arginine group,magnesium sulfate group and(L-arginine+magnesium sulfate) group can lower the mouse blood pressure and urine protein levels of hypertensive disorder complicating pregnancy,prompt NO synthesis,adjust TGF-β1 and IGF-Ⅱexpression, improve uteroplacental circulation,improve fetal and placental development,and lower rates of fetal death,wherein the curative effect of (L-arginine+magnesium sulfate) group is the most significant.
