Objection: Hypertensive disorder complicating pregnancy is a kind of disease that severily harm the health of pregnant women and infants.It is the main cause of pregnant women and infants'death. Althought there are many hypothesizes ,people can not satisfactly interprete the pathogenesy of this diease. Because there is no satisfact therapy for this diease,it is very impormtent that to screen high risk gravida and monitori high-risk group as early as possible.But up to now there are still not specific biomarker in serum can as early scanning indicatrix. Proteomics give us a new method and strategy to study the pathogenesy and the method of early scanning hypertensive disorder complicating pregnancy. Proteomics describes the set of all proteins in a cell, tissue, or organism, proteomics aims ambitiously to study not only protein expression profiles but also protein functions, which should provide researchers with a more comprehensive view of the molecular machinery that governs biology. This type of study requires several data and, thus, the development of both high-throughput technologies for protein investigation and adequate bioinformatics tools for analyzing large data sets.There are many relative facters have effect on proteomics occupying on metabolism dynamic state .People successly make use of proteomic technology not only found many diseases and cancers special biomarke such as,Alzheimer's disease, liver cancer, lymphoblastoma, esophageal carcinoma, ovaries cancer and so on.,but also get many gratifying breakthroughs in pathogenesy of many diseases. Our study apply proteome echnique to scan the serum special biomarker of hypertensive disorder complicating pregnancy.We hope we can get the serum special biomarker of hypertensive disorder complicating pregnancy and some specificness associated proteins of this disease and give us some enlightenment of pathogenesy. Method:1.we select 21 patients that are hypertensive disorder complicating pregnancy and 21 normal pregnancies.We draw their serum mass spectrum with the surface-enhanced laser desorption/ionization time of flight-mass spectrometry to select Specific Biomarkers of hypertensive disorder complicating pregnancy. 2. We gather serum of health pregnancies which during midtrimester pregnancy (13~16 weeks). Tracing these pregnancis'gravid consequence, on the basis of gravid consequence,we divide thes pregnancis into two groups,one is hypertensive disorder complicating pregnancy group ,the other is normal pregnancy group. We draw and analyze the serum mass spectrum of two groups (13~16 weeks) with the surface-enhanced laser desorption/ionization time of flight-mass spectrometry. Secend ,we draw draw and analyze the serum mass spectrum of normal pregnancy different week to select early pregnancy special biomark of hypertensive disorder complicating pregnancy. Result: 1. In the rang of M/Z from 1000 to 20000 there are distinction between two groups'serum mass spectrums. In the serum of hypertensive disorder complicating pregnancy group,the relative amount of protein which M/Z is 3245(0.46±0.41)is obviously lower than normal group(1.28±0.44)(P< 0.05 ), the relative amount of proteins which M/Z are 1324 and 13749(1.59±0.08,0.87±0.03)is obviously higher than normal group(1.23±0.03,0.55±0.07)(P< 0.05 )。2. During the period from 13 to 16 week, in the rang of M/Z from 1000 to 20000,there are obviously different between hypertensive disorder complicating pregnancy group and normal group. there are five proteins'relative amount are obviously different.In the serum of hypertensive disorder complicating pregnancy group the proteins which M/Z are 3942,8895,1324,13749 relative amount are obviously higher than normal group (P<0.05). the proteins which M/Z are 2824,3245 relative amount are obviously lower than normal group(P<0.05).3. Contrast different week serum mass spectrum of normal pregnancy:In the rang of M/Z from 1000 to 20000 ,there are many relative amount of proteins are differences between different weeks.In the serum mass spectrum of midtrimester pregnancy,there are six proteins which relative amount are obviously higher than latterly trimester which M/Z are 2824,4652,6117,2959,6203,5072(P<0.05), though there are eight proteins which relative amount are obviously lower than latterly trimester which M/Z are 2732,9720,4161,4182,8942,9415,3821,4495(P<0.05). Conclusion: 1. During late pregnancy,there are obviously difference in serum mass spectrum between hypertensive disorder complicating pregnancy and normal pregnancy in the rang of M/Z from 1000 to 20000.The proteomics which is make of by three proteins which M/Z are 1324,3245,13749 is the special biomarker in serum of hypertensive disorder complicating pregnancy. 2. In the rang of M/Z from 1000 to 20000,during the period from 13 to 16 week,there are obviously different between hypertensive disorder complicating pregnancy group and normal group. there are five proteins'relative amount are obviously different.This proteomics possible is the special biomarker in serum of hypertensive disorder complicating pregnancy. 3. This scanned proteomics are possible the associated proteins of hypertensive disorder complicating pregnancy that give us new strategto study the pathogenesy of hypertensive disorder complicating pregnancy.
