Effects Of Bushen Tongqiao Decoction On Expressions Of Bax And Bcl-2 In Hippocampal Neurons Of Diabetic Rats

Objective To observe the effects of Bushen Tongqiao decoction on expressions of Bax and Bcl-2 in hippocampal neurons of diabetic rats and to research the therapeutic effect of Bushen Tongqiao decoction on the degeneration of hippocampal neurons of diabetic rats.Methods 40 rats were randomly divided into 4 groups:normal control group,diabetic mellitus control group,traditional Chinese medicine prevention and cure group and western medicine control group.The diabetic rat models were induced by intraperitoneal injection 55mg/kg of streptozotocin(STZ).The rats in traditional Chinese medicine prevention and cure group were drenched with 16g/kg Bushen Tongqiao decoction since 1 week ago before the day STZ was injected,western medicine control group therapy by insulin since the day STZ was injected,while the rats in other groups were drenched with equal liquid normal solution.At the same time of every-week,the weight and fasting blood-glucose of the rats were detected.40 rats were killed for 4 weeks later,fixative was injected intravascularly into the brain and brain tissues were picked up to make into continual paraffin wax sections.The Bax and the Bcl-2 expression level was evaluated by immuno-histochemistry SABC.Totaling the number of the positive expressions as their positive cell's density by objective micrometer. the statisti test was adopted to compare the difference between groups.Results①Compared with normal control group,reflex were slow,the weight decreased and the fasting blood-glucose increased of rats in diabetic mellitus control group(P＜0.01),the positive cell's density of the expressions of Bax increased in hippocampal CA1 and CA3 areas of diabetic mellitus control group(P＜0.05),while the positive cell's density of the expressions of Bcl-2 decreased(P＜0.05)②Compared with diabetic mellitus control group,reflex were quick,the weight increased and the fasting blood-glucose decreased of rats in traditional Chinese medicine prevention and cure group(P＜0.01),the positive cell's density of the expressions of Bax decreased in hippocampal CA1 and CA3 areas of traditional Chinese medicine prevention and cure group(P＜0.01),while the positive cell's density of the expressions of Bcl-2 increased(P＜0.01)③Compared with western medicine control group,the positive cell's density of the expressions of Bax decreased in hippocampal CA1 and CA3 areas of traditional Chinese medicine prevention and cure group(P＜0.05),while the positive cell's density of the expressions of Bcl-2 increased(P＜0.05).Conclusions It is suggested that Bushen Tongqiao decoction is effective in preventing and treating the degeneration of hippocampus neuron of diabetic rats induced by STZ.It might be related to it can reduce fasting blood-glucose and inhibit diabetic rats neuronal apoptosis.Subject word Bushen Tongqiao decoction,diabetic mellitus,rats,hippocampus,Bax,Bcl-2...
