Part I Expressions and clinicopathological significance of MCM5 and E2F-1 in patients with gastric carcinomaObjective:Gastric cancer is one of the most common malignant tumors in China, the morbidity and mortality are high, the abnormal proliferation of cell has important significance in the cancer of the stomach formation and progresses. MCM5 is a member of the minichromosome maintenance proteins, is a kind of discovered in recent years to better reflect the degree of target cell proliferation. E2F transcription factors is one of the important link in cell cycle regulation.This study to explore the correlation between expressions of MCM5 and E2F-1 and clinicopathological parameters of gastric carcinoma was analysed.Methods:Immunohistochemistry SP method was used to detect the expressions of MCM5 and E2F-1 in gastric carcinoma tissues of 57 cases of gastric carcinoma and in 20 cases of normal gastric mucosa. The correlation between expressions of MCM5 and E2F-1 and clinicopathological parameters of gastric carcinoma was analysed.Results: 1. Expressions of MCM5 and E2F-1 in gastric carcinoma and normal gastric mucosa.For the result of the immunohistochemical staining positive signal of MCM5 expression wes located in the nucleus as indicated by a yellow-brown staining.The positive signal of E2F-lwas predominantly marked on the cytoplasm, a few can simultaneously appeared in the nucleus.The positive expression rate of MCM5 in cancer tissues was 71.9%(41/57), while in normal mucosa it was 0%(0/20).The difference between the two groups was statistically significant (P<0.001). The positive rates of E2F-1 in normal and gastric carcinoma tissues were 35%(7/20) and 66.7%(38/57), denoted a significant diference (P<0.05).2. Relationship between expressions of MCM5 and E2F-1 and clinicopathological parameters of gastric carcinoma patients.MCM5 and E2F-1 in cancer tissues were not correlated with sex, age, the cell differentiation, the depth of invasion, lymph node metastases, distant metastases and TNM stage (P>0.05).3. Correlation between MCM5 and E2F-1 expressions in gastric carcinomaThe Spearman correlation analysisest was used to analyze the correlation between MCM5 and E2F-1 expressions in gastric carcinoma, the results show that MCM5 and E2F-1 is closely related (r=0.635,P=0.000).Conclusions:Overexpression of MCM5 and E2F-1 correlates with occurence and development of gastric carcinoma.The correlation between MCM5 and E2F-1 expressions in gastric carcinoma was highly significant positive. MCM5 and E2F-1 may work in the early phase in carcinogenesis of gastric carcinoma.MCM5 and E2F-1 is closely related,to joint detection the two kinds of protein expression may become a molecular marker of gastric cancer diagnosis. Part II Expression and clinicopathological significance of MCM5 in human gastric carcinoma, precancerous lesion and normal gastric mucosaObjective:To investigate expression of MCM5 in human gastric carcinoma, precancerous lesion,normal gastric mucosa and its clinicopathological parameters.Methods:Immunohistochemistry SP method was used to detect expression of MCM5 in 57 cases of gastric carcinoma,32 cases of precancerous lesion and 20 cases of normal gastric mucosa.Results:The positive expression rates of MCM5 in gastric cancer,precancerous lesion and normal gastric mucosa were 87.7%(50/57),78.1%(25/32) and 0%(0/ 20),respectively.The positive expression rates of MCM5 in gastric cancer and precancerous lesion were higher than that in normal gastric mucosa(P<0.01).The strong positive expression rates of MCM5 in gastric cancer and precancerous lesion were 66.7%(38/57)and 25.0%(8/32),the difference was significant(P<0.01).The total positive expression rate of MCM5 in gastric carcinoma tissues was not correlated with cell differentiation,depth of invasion,lymph node metastases andTNM stage (P>0.05).However,the strong positive expression rate of MCM5 in well-moderately gastric carcinoma was less than poorly differentiated gastric carcinoma(P<0.05).Conclusion:MCM5 is a reliable marker of cell proliferation.Increased expression of MCM5 not only indicates the proliferation of malignant cells, but also indicates that the proliferation of underlying malignant cells.Therefore,MCM5 can serve as an effective marker for the diagnosis of precancerous lesion and gastric carcinoma.
