Objective:To investigate the plasma concentration of visfatin in newly diagnosed type 2 Diabetes(T2DM) and normal glucose tolerance(NGT) and its correlationship with the risk factors of type 2 diabetes.2.To investigate the genotype distribution of visfatin -1535 singlencleotide C/T polymorphism and its correlation with insulin resistance index in NGT and T2DM.Methods:1.162 Subjects with normal glucose tolerance(NGT),153 with type 2 diabetes were studied.Plasma visfatin was measured by ELISA assay.BMI,WHR,FBG and insulin and HOMA-IR were measured. Besides,plasma visfatin concentration was compared between the obese and lean subjects(cut point was BMI=25kg/m~2).Linear correlation analysis and multiple stepwise regression analysis were conducted to investigate the correlation between visfatin and risk factors.2.162 NGT and 153 T2DM were genotyped for the polymorphism of visfatin by PCR-RFLP assays.Results:1.The plasma concentration of visfatin in NGT group was 29.00 ng/ml and was 32.47 ng/ml in newly diagnosed T2DM group. Mean fasting plasma visfatin was significantly higher in the type 2 diabetes group in comparison with NGT.In comparison with the male,the plasma visfatin in female was significantly higher than male.There was no significant different between the rest subgroups. Plasma visfatin in obese was significantly increased than that in the non-obese individuals.There was a significant positive correlation between visfatin and BMI,WC,WHR,HOMA-IR,HbA1c.Stepwise regression analysis showed BMI and HOMA-IR were the independent factors for visfatin.2.The was a SNP polymorphism in Zhenjiang Chinese population.The TT,TC,CC genotype frequencies of visfatin -1535 nucleotide T/C polymorphism were 33.3%,39.2%,27.5%respectively. The allelic frequencies were 33.3%,35.2%,31.5%.There was no significant difference about SNPs-1535C / T genotype frequency and allele frequency distribution between the two groups(P>0.05).(2) In T2DM group,high-density lipoprotein cholesterol level was lower in TT genotype than that of other genotypes(20.48±4.82mmol / L for TT vs. 14.94±6.30 mmol / L for TC,P = 0.01),and the 2hPG level was higher than that of other genotypes(20.48±4.82mmol/L for TT vs.14.94±6.30 mmol / L for CC.P = 0.01).Conclusion:1.In summary,we found that plasma visfatin level was significantly higher in type 2 Diabetes.Our conclusion was that there was a coexistance of the increased plasma visfatin with insulin resistance of type 2 diabetes.2.The polymorphism of visfatin -1535 nucleotide C/T polymorphism was associated with glucose and lipid metabolism in Chinese T2DM.
