â… ,Study On Purification Of Silybin And Pharmacodynamics Of Silymarin Solid Dispersion â…¡,Study On Molluscicidal Activity Of JJ18 Inoculated To Beach Soil

There are two parts in the thesis:â… Study on purification of Silybin and pharmacodynamics of Silymarin solid dispersionSilymarin extracted from Haplopappus gracillis Silybum marianum L.is a kind of flavonolignan,it is constituted by silybin,isosilybin,silidanin,silychrison,four kinds of isomers.As a durg to protect liver,Silymarin has accumulated experience for more than 30 years.In order to enhance the bioavailability,we prepared silymarin-PVP solid dispersion and studied on recrytaUization purification.The main content is as follow:(1) The heat reflux mothod was used to extract silymarin from Silybum marianum L.cod,the optimal extraction technology was with acetic ether 6 times for 8 h,78℃.For one-time extraction,the yield of silymarin is 4.69%,for four-time extraction,the yield is 6.18%.(2) We prepared different proportion Silymarin-PVP solid dispersion.The proportion of silymarin and PVP was optimized by the determination of apparent solubility and dissolution,the result was PVP 7 times Silymarin.The apparent solubility of silymarin from solid dispersion is 62 times silymarin in water.After 20 min in artificial gastric juice,the dissolution is exceed 80%.(3) Mouse model of liver injury was set up by giving CCl₄ abdominally,the liver coefficient and the content of ALT,AST,SOD,MDA were examined,which were compared with pellet,interclass aggregated consideration was analyzed by Hism function.It was displayed,when the proportion of silymarin and PVP was 1:7,the solid dispersion could significantly improved five index(liver coefficient, GPT,GOT,MDA,SOD).And pharmacodynamic action of high dose group solid dispersion was close to control group,that meaned the therapeutic efficacy of solid dispersion was better than pellet.(4) Take silymarin as feed,put feed in the 95%ethanol under 80℃,lay up in 4℃refrigerator,we had done recrystallization experiment of Silybin.The recrystallization solvent was 70%ethanol optimizated by purity and recovery of silybin.At that time,the purity of Silybin is above 97%.â…¡Study on Molluscicidal Activity of JJ18 Inoculated to Beach SoilSchistosomiasis is an old local parasitic disease,which has greatly threatened human health acutely.Water-snail is the only intermediate host of blood fluke,so eliminating water-snail has important significance.Endophyte JJ18 had good molluscacidal activity which was obtained from Pseudolarix kaempferi Gord.In order to study the feasibility of killing snail on site,we inoculated JJ18 to the beach soil from breeding ground for snails in the laboratory,and fermented the soil leachate.The survival conditiong of JJ18 is determinded by molluscacidal activity of broth.The main content of the thesis is as follow:(1) The fermentationg products of JJ18 was freeze-dried and extracted by n-butyl alcohol,it turned that intracellular products had no antiblastic effect.Inversely,the exocellular products had bacteriostasis effect on Escherichia coli and Staphylococcus aureus,and the MIC is 0.9 mg/mL and 1.35 mg/mL.(2) JJ18 was inoculated to the beach soil,in order to inspect molluscacidal activity of fermental product of soil leachate.Endophyte JJ18 could survive in soil for at least 30 days.(3) The acute toxicity experiment was done by brine shrimp.The acute toxicity of fermental products of JJ18 inoculated to beach soil is lower than JJ18 inoculated to PDA,the result indicated inoculation had good effection.
