In our paper, silybin was selected as the model of hydrophobic molecule. according to the characteristics of human gastricintestinal fluid, adopting to solid-liquid equilibrium method, the impact of pH and cholate sodium on Solubility of silybin was studied.1.The physical and chemical parameters such as melting enthalpy and solubility of silybin in waters were measured. Experimental results indicated that the solubilities of silybin in waters were small and dissolved speed was slow. And the solubilities of silybin increased adapting to temperature increased. According the relation between solubility and temperature, by using of thermodynamic Properties, the solubility data were correlated. The melting enthalpy and the solubility parameter were calculated, and the data were compared with differential scanning calorimeter measurement data. The compare of solubility parameters between silybin and water illustrated the causation that the solubilities of silybin in waters were relatively small.2.The solubilities of silybin in HCl aqueous solution and NaOH-KH2PO3 buffer solution at different temperature and pH values were mensured. Experimental results shown that the solubilities of silybin were increased with temperature and pH increased, and the effect of temperature was weakened with pH creased. According to Henderson-Hasselbalch (HH) equation, the ionizing constants were calculated, which shown silybin could ionizing out [H+] easily. But the solubilities of silybin were too small, the aqueous solution of silybin ionized out [H+] very small and could not show obvious acidity.3.The solubilituies of silybin in cholate sodium aqueous solution at the different temperature was measured. Experimental results shown that the solubilities of silybin were increased with the increase both in cholate sodium's concentration and temperature.By using of thermodynamic properties, the transfer Gibbs free energy(?trG), enthalpy(?trH) and entropy(?trS) were calculated.
