Objective: To observe the different Tongluo Chinese herbs for diabetes sciatic nerve expression of VEGF. Method: STZ-induced rat model of diabetes were randomly divided into the normal control group (NG), the control group (MG), Xin Wei Tongluo Group (XG), Insects Tongluo Group (CG), Rattan Tongluo Group (TG), the normal control group and control group were given distilled water gavage, Xinwei Tongluo group, Insects Tongluo group and rattan Tongluo group were corresponding to Tongluo herbs gavage. Observation of the rats in the 2 w, 4w, 8w, VEGF immunohistochemistry and Western blot expression and observation of the sciatic nerve morphology and ultrastructural change. Results: Light microscopy showed that NG Group nerve fibers form regularly, distribution orderly, myelin forms dense rules; MG 8w nerve fibers are disordered, myelin density uneven, some of demyelination, a cavity-like changes in the shaft Sudden swelling; three treatment groups at 8 w returned to normal patterns. Immunization show that NG Group sciatic nerve fibers Schwann cells and axons no express or weak positive. MG4w Group sciatic nerve Schwann cells and axons are strong positive expression; MG8w Group sciatic nerve Schwann cells and axons of moderate intensity are positive. Expression of Three treatment group (TG, CG, XG) increased in four weeks, but slightly lower than no treatment group (MG); the ion, slightly lower than no treatment group (MG); 8 weeks was significantly lower than the untreated group. Western blot test results show, 4w, MG, XG, TG and CG express increased (P <0.05)than NG, and the strongest expression is MG (P <0.01); MG, XG, TG and CG expressed more in 8w than NG in 4w. Electron microscope showed that normal rats myelinated nerve fibers myelin and axons structure clear, MG rats significantly change the demyelination, the treatment group myelin resume clear-layer structure, increasing in the number of mitochondria within the axon. Conclusion: VEGF in the nervous system can increase the survival rate of Schwann cells, promote nerve regeneration, blood vessel formation in nerve. Tongluo three different Chinese herbs can repair the sciatic nerve myelinated nerve fibers, so as to diabetes prevention and treatment of peripheral neuropathy provides a new idea.
