ObjectiveDiabetes Mellitus is a chronic disease caused by the disturbance of glucose,fat and protein metabolism.It is generally acknowledged that diabetes was associated with and genetic,environment,viral infection,fat,race,nutrition metabolization imbalances and endocrine disorders.Diabetes Mellitus have become the third serious disease that threaten seriously mankind health behind heaert-cerebrovascular and cancer.Though we can select different kinds of drugs to treat diabetes mellitus,oral hypoglycemic drugs have many adverse reaction and toxic effects.The adverse reaction and Smoggy caused by vein injection insulin for a long time make us to find a better drug in Chinese traditional medicine.Thus it is becoming a focus in medical research to original hypoglycemic agent in Chinese traditional medicine.According to the recent study about hypoglycemic effect of the plant polysaccharides and its mechanisms,it indicates that the hypoglycemic effect of the plant polysaccharides is due to several mechanisms,including the plant polysaccharides can promote the effect of hypoglycemic hormone,adjust the content of hepatic glycogen,preserve islet cells,promote the utilization of the blood glucose by the periphery tissue and organs,meanwhile,depress the effect of hypoglycemia hormone, regulate the activity of glycometabolic enzyme.At present,the mechanism of the treatment of diabetes with fucoidin were seldom reported.Our study reported the effect and the mechanism of the fucoidin on blood glucose in a diabetic mouse model.Further study discussed the influence of the fucoidin in the blood glucose of normal mice and the hypoglycemic effects even the blood lipid metabolism of diabetic mice induced by STZ.Materials and Methods1.The effects of the fucoidin on the normal mice20 mice were divided into 2 groups randomly as the control group and the experimental group.Control group were fed with the volume of distilled water,and experimental group were fed with fucoidin(500mg/kg).The blood glucose were determined at the 14th days while the mice were absolute diet and without absolute water the night before.2.The effects of the fucoidin on the diabetic mice induced by STZ30 mice were divided into 3 groups randomly as the model group,fucoidin group and the positive group.Another normal 10 mice were as negative group.All the mice were lavaged every morning,however,the fucoidin group were lavaged with fucoidin (500mg/kg),and the model group and the negative group were lavaged with the volume of distilled water,but the positive group were lavaged with oral hypoglycemic drugs dimethyldiguanide(160 mg/kg).Continuous treatment for 32 days,then the blood glucose,the TG and TC in the serum were determined at the 8th,21th,32th day while the mice were absolute diet and without absolute water 12 hours before.3.Statistical analysisStatistical analysis were used SPSS software,and each group means were showed with x±s,compared between groups were used variance analysis.Results1.Fucoidin(500mg/kg) had not significantly influenced the fasting blood glucose and lipid of normal mice(P>0.05).2.In the diabetic mice induced by STZ2.1 Compared to model groups,at the 32th day after treatment,the mice blood glucose of the fucoidin and the positive(oral hypoglycemic drugs dimethyldiguanide) groups were significantly decreased(P<0.05,P<0.01).2.2 The TC and TG of the mice serum has no significantly difference between the control and the fucoidin groups.ConclutionFucoidin has a effective hypoglycemic activity on diabetic mouse model,and fucoidin is a very promising polysacchafides hypoglycemic drugs.
