Objective: To study the effect of GHRP on ET-1 and IL-1βin the course of CHF, meanwhile explore the function on the period of growth and development for purpose of providing better theoretical base in clinic prevention and treatment.Methods: 60 healthy Wistar female rats are selected and divide randomly into three groups which are normal group(group A), model group(group B)and treatment group (group C), and 20 in each group. We set up the CHF models by injecting ADR ,group C is used GHRP to treat. we use:①Useful methods such as monitoring the hemodynamic change and HE dyeing of myocardium of rats to judge whether these rats suffer from CHF;②Electron microscope to observed morphological changes of myocardial cells;③IHC method to examine the change of the form and density of immunological positive cell of ET-1 and IL-1β;④PCR technology, the myocardium of rats in each group to analyze the expression of ET-1 and IL-1β;⑤ELISA method to determine the changes of the contents of ET-1 and IL-1βin myocardial tissue of rats.Results:①Compared with group A, the changes of hemodynamics and myocardial of group B were significant, and the rats of group B suffer from CHF; But group C has no obvious change;②Group A keep normal mitochondrial structure, and intact of intercalated disc structure; group B has mitochondrial vacuolar degeneration in a few regions, intercalated disc structure had no change; there appeareed mitochondrial swelling, and vacuolar degeneration, and intercalated disc lower electron density in many regions;③Immune-positive cells in group A scattered, and nucleus located in the central; immune-positive cells in group B tend to be hypertrophy, many nucleus stay away from center at the side, and Gather distributed; immunopositive mast cells in group C reduced significantly, many nucleus stay in the central , cells scattered;④mRNA expression levels of ET-1 and IL-1βin myocardial tissue of group B are higher than that of group A and C obviously; mRNA expression levels of ET-1 and IL-1βof group C are a little higher than that of group A , and they have no obvious difference;⑤The contents of ET-1 and IL-1βin myocardial tissue of group B are higher than that of group A and C obviously(P<0.05); The contents of ET-1 and IL-1βof group C are a little higher than that of group A , and they have no obvious difference(P>0.05).Conclusion:①The contents of ET-1 in myocardial tissue can be degraded by GHRP;②The contents of IL-1βin myocardial tissue can be degraded by GHRP;③GHRP can improve myocardial remodeling in the course of CHF, GHRP reduces the expression of ET-1 and IL-1βin myocardial tissue in the course of CHF is possibly one of ways.
