Objective:To determine the effects of BPP on osteoporosis in rats,the BMD and relative biochemical molecules were meatured.Methods:The experimental model of osteoporosis(OP) induced by dexamethasone (DXM) in rats were performed in this study.And the effect of tibia defect healing in rats was also observed.(1) 60 of 3-month-old Wistar rats were randomly divided into 6 groups:the negative control group(water,10 ml/kg/d);the model group(DXM, 2.5mg/kg);the positive group(Jiegu Qili Pian,300mg/kg);the high dose group (BPP,200mg/kg);the moderate dose group(BPP,100mg/kg);the low dose group (BPP,50mg/kg).In addition to the negative control group,the other groups injected dexamethasone(2.5mg/kg),twice a week,for 6 weeks caused the model of osteoporosis.After 12-week period continous,the urinary samples of all rats were collected before they were killed,and then,the blood samples were collected.The samples of urine and blood were taken for determinination of serum calcium(Ca), phosphorus(P),alkaline phosphatase(ALP),bone glaprotein(BGP),urinary calcium (Ca),creatinine(Cr),phosphorus(P) and urinary hydroxyproline(HOP).The bone mineral density(BMD) of the rats in all groups were determined.After executed,kept back the tibia for making decalcified slice,and then for the analyse of bone tissue histomorphometry.(2) 100 of 3-month-old Wistar rats made of the tibia bone defect model were randomly divided into 5 groups:the control group(water,10 ml/kg/d);the positive group(Jiegu Oili Pian,300mg/kg);the high dose group (BPP,200mg/kg);the moderate dose group(BPP,100mg/kg);the low dose group(BPP,50mg/kg);20 rats in each group.For 20 days after administeration of BPP, half of rats tibial were taken for radiology observation and callus histology.40 days after administeration,the remainder rats tibial were taken for radiology and callus histology.SPSS 13.0 statistical package was used to deal with all the data.There was significant difference when P<0.05.Results:The concentrations of serum phosphorus(P) in the high,moderate,low dose group were 2.28±0.35 mmol/L,2.34±0.32 mmol/L,2.29±0.30 mmol/L, respectively.It was significantly lower than that in the model group(2.61±0.23 mmol/L)(P<0.05);The concentrations of serum ALP in the three groups were 94.50±19.79 U/L,102.1±23.96 U/L,110.1±23.53 U/L,respectively.It was significantly lower than that in the model group(137.9±23.99 U/L)(P<0.05);The concentrations of serum BGP in the three groups were 1.69±0.27 ng/L,1.80±0.20 ng/L,1.73±0.17 ng/L,respectively.It was significantly lower than that in the model group(1.97±0.24 ng/L)(P<0.05);The concentrations of urinary Ca(Cr)/HOP(Cr) in the three groups were 0.16±0.05 mmol/μmol /0.20±0.05μg/μmol,0.18±0.04 mmol/μmol/0.24±0.05μg/μmol,0.22±0.04mmol/μmol/0.25±0.05μg/μmol,respectively. It was significantly lower than that in the model group(0.27±0.08 mmol/μmol /0.29±0.07μg/μmol)(P<0.05).There was no significantly difference in the concentration of the serum P and urinary P/Cr in the all groups,respectively.The BMD in the model group was 0.136±0.012(g/cm~2),it was significantly lower than that in the negative control group(0.161±0.014 g/cm~2,P<0.05);the values were 0.150±0.015 g/cm~2,0.152±0.015 g/cm~2,0.148±0.013 g/cm~2 in the high,moderate,low dose group,it was significantly higher than that in the model group(P<0.05).The values of the Tb.Th were 48.73±12.27,48.17±10.28(μml) in the high,moderate dose group,it was significantly higher than that in the model group(36.23±12.27μml P<0.05).The values of the Tb.Tr were 44.30±8.98,38.41±11.27,40.59±9.88(%) in the high,moderate,low dose group,it was significantly higher than that in the model group(29.57±7.19%P<0.05).The bone trabecular became thinner,and arranged in disorder in OP rats,however,the status was reversed obviously by administrating BPP. (2) 20 days after administeration,the radiology results showed that in the three-dose BPP group bone defect line healed faster than in the blank group;and the histology showed that the osteoblasts were more active in the BPP group than in the blank and bony trabecula in the BPP in group were more mature than those in the model group. 40 days after administeration,the radiology results showed that in the high and moderate dose BPP group bone defect line almost complete healed but in the model group still visible;and the histology showed that the bone trabecular of the callus in the BPP group is orderly,but in the model group is disorderly.Conclusion:BPP improves some osteoporosis symptoms in rats by increasing bone formation gradually,inhibiting bone resorption and accelerating bone remodeling. Moreover,BPP was showed an anti-osteoporosis action in rats.
