ObjectiveIn this project, we investigated the effect of Pingyangmycin (PYM) on epithelia of odontogenic keratocyst (OKC) through detecting expression of keratins for preparation to explore a new method to cure OKC before in vivo and clinical study.MethodsWall specimens were collected from twenty OKC patients who had been diagnosed in Department of Oral & Maxillofacial Surgery in Qilu Hospital of Shandong University from Jan. to Dec. in 2008. Specimens were cut into pieces and cultured under designed circumstances, and then treated with 10μg/mL, 100μg/mL, 1000μg/mL PYM or without PYM as controls. Immunohistochemistry (IHC) was used to detect the expression of keratins by clones AE1/AE3, which is a pan-keratin antibody, in epithelial tissues of OKC after 2h, 24h or 48h (Oh as controls). The positive rates and expression intensities were calculated and recorded by average±standard differentiation ( x±s ). Then the differences of the data between groups were analyzed by SPSS16.0 statistics software.ResultsThe expression intensities of keratins showed statistical differences (P<0.05) among groups with different concentrations, and the differences between each two groups with different concentrations showed statistical significance (P<0.05), as the average compare showed, the higher concentrations, the less expression intensities of keratins were. The expression intensities of keratins showed statistical differences (P<0.05) among groups with different durations, and the differences between each two groups with different durations, showed statistical significance (P<0.05), as the average compare showed, the longer duration, the less expression intensities of keratins were. Furthermore, the concentrations and durations of PYM were found to display interactive effects (P<0.05).ConclusionsThe expression intensities of keratins showed statistical differences among groups with different concentrations and time. And with higher concentrations and more time, the expression intensities of keratins were less. As the expression of keratins could represent the extent of depression of epithelia, we can conclude that PYM showed effect of depression on epithelia of OKC.
