The Empirical Study Of The Influence Of Fugu Jianbu Pellet On Rabit's Chondrocyte Apoptosis Resulted From Articular Genu Arthritis

1.Objective:Observe the the influence of Fugu jianbu pellet(notoginseng, Lycium chinense Mill.,danshen root,asiatic cornelian cherry fruit,gryllotalpa, astragali,radix,rehmannia dride rhizome,ramulus cinnamomi,common bletilla tuber) both on rabit's chondrocyte apoptosis resulted from articular genu arthritis and on the genetic expression of Bax,Bcl-2.Approach the therapeutics mechanism of Fugu jianbu pellet with articular genu arthritis2.Means:devide 48 rabits into 4 groups by random,A for normal control group, B for analog group,C for zhuangu gujie pellet group,D for fugu jianbu pellet group,Rabbits in B,C and D group were established into Keen Osteoarthrisis models by plaster fixation on the knee of the left leg for 6 weeks.Rabbits in A and B group were lavaged 10ml sodium chloride with no drugs in it;Rabbits in C group were lavaged 10ml sodium chloride with 0.4g Zhuangguguanjiepill,twice and in D group were lavaged 10ml sodium chloride with 0.756g FuGuJianButablet,a day.Then,kill 4 Rabbits in each group in 2weeks4weeks and 6weeks and take the articular cartilage to observe:1. observe the cartilage cell apoptosis and counted number adopting the model of in situ end labelingz;2.Detect the expression level of Bax,Bcl-2 by immunology-histochemical method. 3.Results:1).the detection of rabit's chondrocyte apoptosis in situ end labeling(TUNEL): in the 2 week,the Apoptosis rate of groupC,D are significantly infra group B(P＜0.01),it has no significant deviation between group C and group D;in the 4,6 week,the Apoptosis rate of group B,C,D are all significantly super the group A,groupD are significantly infra group B,C(P＜0.01);comparing between the 2 weeks,4weeks,6weeks of group C,the results are all significant difference(P＜0.01).2).detectong of the gene(Bcl-2) expression about abaissement apoptosis:The results of group D are all significantly super group B,Cand infra group A in the 4,6 week,variance is significance(P＜0.01).3).detectong of the gene(Bax) expression about encourage apoptosis:The results of group D are significantly infra group B,C(P＜0.01) in the 4 week.it has no significant deviation between group C and group D in the 6 week,not statistically significant(P＞0.05).4.Conclusion:1).fugu jianbu pellet conspicuously helps depress the apoptosis rate of cartilage cell suffering from hypertrophic arthritis,to achieve the protection cartilage cell and delay the onset of hypertrophic arthritis.2.) fugu jianbu pellet conspicuously helps depress the apoptosis rate of cartilage cell,the possible mechanism is that it helps prevent the expressing of gene Bax and conspicuously enhance the expressing of Bcl-2 in hypertrophic arthritis.
