Objective:To investigate the function of the Fluid of Jianbu Tongluo Fumigation on Mitogen-activated protein kinases which includes p38 MAPK,c-Jun and ERK in articular cartilage of Rabbits knee osteoarthritis,and discuss the effect of the Fluid of Jianbu Tongluo Fumigation on articular cartilage.Methods:4~6months twenty six New Zealand white rabbits were chose, half male and half female. Six rabbits of them were randomly selected as blank group, and the twenty of them were made into knee osteoarthritis by modified Hulth method. After four weeks, the two of them were randomly chose to verify that the mold was successful by hematoxylin-eosin staining and pathological changes through microscope. Then eighteen rabbits were randomly divided into model group, fumigation group and calcitonin group. The rabbits in Fumigation group were fumigated with the fluid of Jianbu Tongluo fumigation one at a time. Calcitonin group were cured with calcitonin intramuscularly one time a day. After twenty days, all rabbits were sacrificed. After that, I would observe rabbit’s general configuration of articular cartilage by eyes and microscope. Last, using PCR technique to detect the expression of ERK1/2、p38MAPK、JNK.Results:(1) General morphological observation: normal group articular cartilage’s appearance, color and surface were normal, and subchondral bone wasn’t exposed. Model group’s articular surface is rough, the color is dark, articular cartilage was exposed and a large number of edge numerous osteophyte were formed; Calcitonin group and fumigation group, cartilage surface were less than normal group, individual areas’ color were dark, and we can see a little of osteophyte.(2) HE staining: by Mankins scores, model group’s cartilage structure, articular chondrocytes and tide line were very different compared with normal group, the damages was very serious, the differences were statistically significant( P<0.05).The damages of fumigation group and calcitonin group were more serious than blank group. However the damages were less than blank group. So the treatment of the two groups can protect articular cartilage. The results of fumigation group and calcitonin group have no difference(P>0.05).(3) PCR Detection: Compared with the normal group, the express of p38, ERK1/2 and JNK of other groups increased significantly(P<0.05); the fumigation group and calcitonin group were lower than model group, the difference was significant(P<0.05); and the differences between fumigation group and calcitonin group, has no significance(P>0.05).Conclusions:(1)We can found that the Fluid of Jianbu Tongluo Fumigation can affect the expression of p38, ERK1 / 2 and JNK through MAPKs pathway. Then it can intervene articular cartilage of osteoarthritis pathological changes, and protect the articular cartilage, delay the OA progress.(2) Through the data of fumigation group and calcitonin group, we can draw the following conclusion that the Fluid of Jianbu Tongluo Fumigation and CT have a certain effect on OA.
