ObjectiveObserving the expression and change of CyclinD1 and p16 in the vitro cultured HaCaT cell line after stimulating by different concentration arecoline,to further gain insight into the carcinogenesis of arecoline.MethodsCultured HaCaT cell line in vitro,then stimulating them by different concentration arecoline for 24 hours.And using the reverse transcription polymerase chain reaction(RT-PCR) to detect the expression of CyclinD1mRNA and p16mRNA.Result(1)In the vitro cultured HaCaT cell line,compared with the control group,CyclinD1mRNA expression was increased by the 40,80μg/ml arecoline groups(P<0.05).(2)In the vitro cultured HaCaT cell line,compared with the control group,p16mRNA expression was decreased by the 20,40,80μg/ml arecoline groups(P<0.05)ConclusionArecoline at certain concertrations can up-regulate CyclinD1 expression and down-regulate p16 expression in the vitro cultured HaCaT cell line,the abnormal expression of CyclinD1 and p16 may be involved in the carcinogenesis of arecoline.
