Significance Of Detection Of PTEN,CyclinD1 And DNA Content In The Carcinogenesis Of Colorectal Mucosa | | Posted on:2010-04-01 | Degree:Master | Type:Thesis | | Country:China | Candidate:F Wang | Full Text:PDF | | GTID:2144360278473845 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | OBJECTIVE:To investigate the expression of PTEN,CyclinD1 and DNA content in the carcinogenesis of colorectal mucosa,and to discuss their relationships.METHOD:The expression of PTEN and CyclinD1 in 58 cases of human colorectal carcinomas,18 cases of adenomatous polyps and 14 case of tumor-side normal tissue were detected by immunohistochemical technique;DNA content was measured by flow cytometry.RESULT:1.The expression of PTEN protein:①Positive immunostaining with the antibody was observed in the cytoplasm of cells.②The positive expressive rate of PTEN in the tumor-side normal tissue,adenomatous polyps and colorectal carcinoma, were 92.86%,83.33%and 65.52%respectively and had a descending trend.The positive expressive rate of PTEN in the colorectal carcinoma was significantly different from that in the adenomatous polyps and the tumor-side normal tissue, (P<0.01).③There was no significant diference between tumor-side normal tissue and adenomatous polyps group(P>0.05),but both were higher than cancer group(P<0.005).④The expression of PTEN in colorectal carcinoma were associated with degree of differentiation of carcinoma,Dukes stage and lymph node metastasis of the tumors(P<0.05).PTEN protein expression had no relation to the patients'age,sex,tumor size and location.2.The expression of CyclinD1 protein:①Positive immunostaining with the antibody was observed in the nuclear.②The positive expressive rate of CyclinD1 in the tumor-side normal tissue,adenomatous polyps and colorectal carcinoma,were 7.14%,16.17%and 60.34%respectively and had a accending trend.The positive expressive rate of CyclinD1 in the colorectal carcinoma was significantly different from that in the adenomatous polyps and the tumor-side normal tissue, (P<0.01).③There was no significant diference between tumor-side normal tissue and adenomatous polyps group(P>0.05),but both were lower than cancer group(P<0.005).④The expression of Cyclin D1 in colorectal carcinoma were associated with degree of differentiation of carcinoma,Dukes stage and lymph node metastasis of the tumors(P<0.05).CyclinD1 protein expression had no relation to the patients'age,sex,tumor size and location.3.The relationship between the expression of PTEN and CyclinD1:There was a negative relationship between the expression of them in the colorectal carcinoma tissue,r=-0.7135,P<0.05.4.The relationship between the index of DNA content and the expression of PTEN and CyclinD1:①The rates of DNA ploidy in the tumor-side normal tissue, adenomatous polyps and colorectal carcinoma were 0,5.56%,68.97%respectively and had a accending trend.②The DNA index(DI) and sphase fraction(SPF) in the positive group of PTEN were lower than those in the negative group,and the DI in the positive group of CyclinD1 was higher than this in the negative group(P<0.05).The aneuploid ratio and SPF in lymph node metastasis positive patients were higher than those in the negative patients(P<0.05).CONCLUTION:1.The abnormal expression of PTEN and CyclinD1 participate the malignant conversion process of the colorectal mucosa cell and correlates with tumor stage, histological poor differentiation and lymph node metastasis;there was a relationship between the expression of them.2.There was a relationship between the index of DNA content and the expression of the PTEN/CyclinD1 in the three kinds of tissues observed.3.The expression levels of PTEN,CyclinD1 and the DNA content may be the important indicator to evaluate the pathologic biology behavior and the prognosis of the colorectal carcinoma. | | Keywords/Search Tags: | Colorectal carcinoma, PTEN, CyclinD1, DNA content | PDF Full Text Request | Related items |
