Aim To study on the hypoglycemic mechanism by sequoyitol and its analogues.Methods Measurement of activitis and kinetic parameters by ultraviolet spectrophotometry of the enzymes,incudingα-amylase,α-galactosidase, trehalase,invertase,α-glucosidase,β-glucosidase,β-galactosidase and glucoamylase,were uesd to detect the inhibition effect in vitro.PNPG was selected as substrate ofα-glucosidase,β-glucosidase andα-galactosidase.ONPG was selected as substrate ofβ-galactosidase. Starch was selected as substrate of amylase and glucoamylase.Sucrose was selected as substrate of sucrose.Trehalose was selected as substrate of trehalase.Glucose tolerance test were used to detect the system effect of the drug in mice.Starch,sucrose or glucose was administered to mice 30min after inositol was administered.Then collect the blood samples at 10,20,30,60,120 min,and measure blood glucose by glucometer.Results Sequoyitol and its analogues show significant inhibiton onα-glucosidase,β-galactosidase and glucoamylase,but not other enzymes detected.It is found that sequoyitol has some inhibitory onα-glucosidase andβ-galactosidase.The IC50 are 0.962 and 23.77 mmol·L-1.Pinitol and myo-inositol has some inhibitory on glucoamylase andα-glucosidase,and the IC50 are 2.4 and 5.30 mmol·L-1 separatly.The inhibitory ratio of Sequoyitol can reduce the postprandial blood glucose in mice. Conclusions The data shows that sequoyitol can decrease the postprandial blood sugar level,part due to its inhibition of glycosidase.
