Objective:To observe the the change of peripheral blood Th17 cells and relationship between the severity and Th17 cells in patients with acute asthma.Methods:Recruit the patients with mild acute asthma(n=10),severe acute asthma(n=10) and healthy volunteers(n=10).The 15ml peripheral blood from the persons were collected and then the peripheral blood mononuclear cells(PBMC) were separated.The PBMCs were maintained at humidified atomosphere of 95%air/5%CO2 at 37℃for four hours in fresh RPMI-1640 culture medium supplemented with 10%FCS(fetal calf serum).And then T-lymphocytes were collected from the PBMCs,number the T-lymphocytes,and then divided them into three groups as following:A group(T-lymphocytes from healthy volunteers);B group(T-lymphocytes from patients with mild acute asthma);C group(T-lymphocytes from patients with severe acute asthma). T-lymphocytes were floated in RPMI-1640 culture medium.Flow cytometer(FCM) were used to detect the expression of peripheral blood Th17 cells.The date were expressed with mean and standard deviation (x±s).Statistic was analyzed with SPSS 11.0 soft-ware.If P is less than 0.05,the results have achieved statistical significance.Results: 1.The result of FCM:The ratio of positive Th17 cells of peripheral blood in the severe acute asthma group(group C)(64.36±4.36)%were more than that in the mild acute asthma group(group B)(40.06±1.75)%(P<0.05) and the ratio of positive Th17 cells of peripheral blood in health volunteers group(group A)(30.72±1.82)%were the least among all groups(P<0.05,respectively)2.Correlation of analysis:The positive Th17 cells of peripheral blood in patients with acute asthma were positive correlation with severity of acute Asthma(r=0.869,P<0.05).Conclusion:(1) The positive Th17 cells of peripheral blood were increased in patients with acute asthma.(2) Th17 cells maybe participate the pathophysiology procedure of acute exacerbation asthma.
