| Objective To detect and compare the expression of MMP-2 and MMP-14 in intervertebral discs of different degeneration stages,to study their contribution in the process of degeneration,to investigate the relationship between the expression of MMP-2,MMP-14 and the degeneration stages of intervetebral discs,analysis their correlations.Methods The tissues of the degenerated discs were obtained from 40 lumbar disc herniation(LDH) patients in the Spine Surgery Department of Xiangya Second Hospital from February 2008 to July 2008.As the control group,20 normal lumbar discs were harvested from 12 patients(7 males,5 females,aged 15-44y),5 lumbar fracture and 7 idiopathic scoliosises.After the discs were obtained,washed by physiological saline, and then fixed and stored in 10%formalin for 24 hours,embedded in paraffin and cut to sections 5.0μm thick.The samples were detected by means of immunohistochemical straining respectively and compare the difference of the degeneration stages by statistical methods.Result1.The positive cell ration of MMP-2 statement observed in degenerated discs is 82.5%,significantly higher than in normal group.2.The positive cell ration of MMP-14 statement observed in degenerated discs is 67.5%,significantly higher than in normal group. 3.A positive correlation was observed between the amount of MMP-2 and MMP-14.Conclusion The expression of MMP-2 and MMP-14 are clearly correlated with disc degeneration,the expression of MMP-2 correlated positively with MMP-14. |
