ObjectiveTo study of signal transducer and activator of transcription3(STAT3) and vascularendothelial growth factor (VEGF) in osteosarcoma tissue expression. Analysis ofcorrelation between the expression of STAT3and VEGF with osteosarcoma diagnosis,histological grade, prognosis and metastasis in order to provide a reference for clinicaldiagnosis and treatment.Method1、50cases of hospitalized patients with osteosarcoma were stuided selected in June2007-between September2009. Tissue microarray and immunohistochemical methodsdetected the expression of osteosarcoma tumor tissue and normal tissue tumors of STAT3,VEGF and CD34.2、The application of Kaplan-Meier survival analysis STAT3, VEGF and survival ofthe time, the difference was significant test application log-rank test methods, and Coxproportional hazards model for survival factors for multivariate analysis.Results1、STAT3in osteosarcoma tissues positive staining localized in the nucleus and/orcytoplasm; pale yellow to dark yellow granular substance. Experiment group osteosarcomaSTAT3expression was78.00%in the control group of normal tissue STAT3expressionwas8.00%, the difference was significant (P <0.05). Experimental group osteosarcomaSTAT3positive area was0.321±0.058, the average optical density of0.155±0.012,control group of normal tissue STAT3were0.283±0.012,0.095±0.004, the differencewas significant (P<0.05)2、VEGF expression in osteosarcoma cells was mainly located in the cytoplasm, paleyellow to brown granular substance. Experiment group osteosarcoma VEGF expressionwas68.00%in the control group without VEGF expression in normal tissues, withsignificant difference (P <0.05). Experimental group osteosarcoma VEGF positive areawas0.416±0.076, the average optical density of0.226±0.053, control group of normal tissue VEGF were0.088±0.007,0.093±0.039, the difference was significant (P<0.05).3、MVD patients with osteosarcoma STAT3expression value of25.72±6.12, negativeexpression was8.07±3.79; MVD expression of VEGF in patients with osteosarcoma valueof40.24±7.98, negative expression was15.55±5.91; patients with osteosarcoma STAT3,VEGF expression and microvessel density was significantly associated (P <0.05).4、 Univariate analysis shew that the expression of STAT3and VEGF withosteosarcoma Enneking classification, the degree of tumor differentiation and distantmetastases, lung metastases, distant tissue infiltration had a close relationship, but with sex,age, pathological osteosarcoma independent of type and location of the tumor. Theseresults were statistically significant (P <0.05). Expression of STAT3and VEGF inosteosarcoma shew a significant positive correlation (r=0.524, P <0.05).5、Kaplan-Meier survival analysis showed that osteosarcoma STAT3and VEGFexpression in patients with higher levels of positive, the shorter its lifetime. Cox hazardmodel analysis shew that STAT3and VEGF expression and MVD was an independentprognostic factor affecting survival of osteosarcoma tumor patients.Conclusion1、Sustained activation of STAT3gene expression in osteosarcoma genesis anddevelopment may play an important role in the clinical diagnosis and treatment ofosteosarcoma.2、STAT3and VEGF expression and osteosarcoma Enneking classification, the degreeof tumor differentiation and distant metastases, lung metastases, distant tissue infiltrationhave a close relationship.3、STAT3and VEGF may participate in the generation process of peripheral vascularosteosarcoma, STAT3and VEGF expression was positively correlated. They play animportant role in the generation and transfer of vessels in osteosarcoma.
