Objective: To comparator and study the pathological change and other interrelated index that effecting by various doses of ephedrine to the heart and kidney. And explore the side effects in rats after cerebral ischemia/reperfusion injury. It is helpful to establish reliable fundamental theory for clinical drug security.Methods: 96 Sprague-Dawley(SD)rats, weighting 220-250g, were used to prepare left middle cerebral artery occlusion (MCAO)models by using modified Longa ligation. Divide the rats into 4 groups randomly: A group for ephedrine, B group for ephedrine, C group for ephedrine, and the blank matched group(D group). Select 4 times(1st week,2nd week,3rd week,4th week)after the operation to observe. The blood pressure and pathological section of heart reflect the situation of cardiac damage. The serum creatinine and pathological section of kidney reflect the situation of kidney damage.Results:⑴The determination of blood pressure showed that all the measured values of medication groups were lower than that of the blank matched group. While the measured values of A group was the lowest and lasts the longest of all.⑵The determination of serum creatinine showed that all the measured values of medication groups were higher than that of the blank matched group. While the measured value of A group was the highest. Furthermore, all the measured value reached to a peak value at the 3rd week whose differences has statistical significance, then began to fall back at the 4th week; (3)The result of pathological section showed that the myocardial fiber gap had widened and there were apparent congestive in the medication groups; and it also showed that the renal glomeruli congestion, vacuolar degeneration and granular degeneration on the renal tubular epithelial cell.Conclusion: In our study, the side effects of 2.0mg/kg/d ephedrine on the heart begin to improve at the 3rd week, while the side effects of 3.0mg/kg/d and 2.5mg/kg/d ephedrine didn't change at the 4th week. The side effects of all doses of ephedrine on the kidney reached to a peak at the 3rd week, and began to fall back at the 4th week. All doses of ephedrine have side effects on the heart and kidney in rats after cerebral ischemia/reperfusion injury. But these side effects are transient, which correlated with the dose. The side effect enlarge with the dose increasion.
