Objective To observe the change of blood plasma level of APN and the expression of ADIPOR and the cardiocyte hypertrophy in non - infarction myocardium after myocadial infarction in rat, and to investigate the effect of rosiglitazon on the blood plasma level of adiponectin and the expression of adiponectin receptor and the cardiocyte hypertrophy after myocadial infarction, to explore the possible mechanism of the benefical effect on the cardiocyte hypertrophy after myocadial infarction.Method 22 adult male Sprague -Dawley rats were subjected to operation of ligation of left anterior descending coronary, The 16 rats surviving for 24 hours after operation were randomly separated into two groups, Myocardial infarction group (M group), Rosiglitazone group(R group),Each group including 8 rats,There was a sham operation group (S group) including 8 rats (The rats were subjected to brading under left anterior descending coronary,not ligation),The rats were growed for six weeks, We got the complete data of 24 rats.Determination of the level of blood glucose before operation and after six weeks,Weigh of the left ventricle, Dermination of cardiocyte area by image analysis and the expression of protein of ADIPOR by Immunohistochemistry (IH) and the blood plasma level of APN by Enzyme linked immunosorbent assay (ELISA).Result (1)Compared with S group,The blood glucose level in M group and R group before operation and after six weeks was not different significantly.(2)Compared with S group,The relative weight of left ventricle and the area of cardiocyte in M group and R group increased siginificantly (p <0.05 ), Compared with M group,The area of cardiocyte in R group reduced siginificantly ( p<0.05) , The relative weight of left ventricle reduced not siginificantly( p>0.05) (3) Compared with S group, The expression of adiponectin receptor 1 (ADIPOR1)and adiponectin receptor 2 (ADIPOR2) in non-infarction mycardium in M group and R group reduced,And the expression of ADIPOR1 reduced siginificantly ( p<0.05) , The expression of ADIPOR2 reduced not siginificantly(p>0.05), Compared with M group, The expression of ADIPOR1 and ADIPOR2 in R group increased, And the expression of ADIPOR1 increased siginificantly (p <0.05 ), The expression of ADIPOR2 increased not siginificantly(p>0.05).(4) Compared with S group, The blood plasma level of adiponectin (APN) in M group reduced siginificantly ( p<0.05) , Compared with M group, The blood plasma level of APN in R group increased siginificantly( p<0.05) ,The blood plasma level of APN in R group is near to the normal level.Conclusion (1)The cardiocyte area increased and the blood plasma level of APN and the expression of ADIPOR1 and ADIPOR2 in non-infarction myocardium reduced significantly after myocadial infarction.(2)Rosiglizatone could inhibit the hypertrophy of cardiocyte and increase the blood plasma level of APN and the expression of ADIPOR1 and ADIPOR2 in non-infarction myocardium without affecting the level of blood glucose. (3)The effect of rosiglizatone on inhibiting the hypertrophy of cardiocyte may correlate with the increasing of the blood plasma level of APN and the expression of ADIPOR1 and ADIPOR2 in myocardium.
