Objective:Stem cells are the the origin cells of other types of cells in the body. With self-renewal, multilineage differentiation and characteristics of steady-state controlOne stem cell can develop into a complete living organisms or tissues and organs. As other cells, breast stem cells is also Multi-potential,they can generate progenitor cells at different level.and differentiate into myoe- pithelial and ductal finilly. Embryonic stem cell gene Oct4 is playing the crucial role in maintenanceing undifferentiation condition and adjusting differentiation process of stem cells,as well as participating the self- renewal of stem cells. So it is the potential useful stem cell marker .Breast cancer as one of the most common malignant tumor,which closely related to abnormal cell cycle regulation. some researchers consider that cell cycle regulation of mammary stem cell disorder is an important mechanism for carcinogenesis. Under normal circumstances. a typical cell cycle phase mainly composed of four phase. Four-phase human cells are regulated by a series of factors.In our research ,to investigate the functional significance and relation- ship of Oct4 and CyclinE in the breast cancer.Methods: Immunohistochemical S-P method was used to detect the expression of Oct4 and CyclinE in human breast cancer.Results: The positive of Oct4 and CyclinE were mostly located in nucleus with rate ofâ… degree15.38% (2/13),â…¡degree 47.06%(8/17),â…¢degree 83.33%(5/6) respectively. The expression levels of Oct4 and CyclinE protein correlated with histological grade (P<0.5). The expression level of CyclinE protein correlated with ER,PR status. There was no significant difference among patiens age, size of tumor and lymph node metastases, and there was no correlation between Oct4 and cyclinE.Conclusions: The over-expression of Oct4 and CyclinE may play a significant role in the incidence and development of breast cancer. Oct4 and CyclinE correlated with the differentiation degree of breast cancer. The expression of Oct4 correlated with ER. Oct4 doesn't correlate with CyclinE. Above all, these factors will be more helpful for treating breast cancer.
