This work was supported by National Science and Technology Supporting Promgram for the 11th Five-Year Plan(2006BAI09B05-1;2007BAI37B02) and "Quality Standards Research for Angelica Sinensis" of "Chinese Pharmacopoeia" (YZ-105) in 2010 version.There were three parts in this dissertation.In the first part,there was one chapter.A detailed review was given to the research progress of Angelica Sinensis including herbal textual research,resources, harvest,chemical constituents,pharmacological effects and quality standard.In the second part,there were three chapters.Studied on the the resource chemistry of Angelica sinensis in different regions and harvest times.In the first chapter,it was studid on the content changes of polysaccharides of Angelica sinensis in different regions and harvest times.The contents of netural polysaccharides and acidic polysaccharides were determined by the sulphuric acid-phenol colorimetric method and the sulphuric acid-carbazole colorimetric method.The content of total polysaccharides was about 14.58~26.76%and it was Sichuan>Yunnan>Gansu in different regions.The content trend was gradually increased from August to September,the highest in October.In the second chapter,it was studied on the dynamic accumulations of Angelica Sinensis volatile constituents in different regions and harvest times.32 compounds were idendified in volatile constituents by the GC-MS method.The results of similarity evaluation and hierarchical cluster analysis showed the components and its relative contents in volatile constituents had higher similarity degrees from different regions in October,but it had larger differences in the same regions from different harvest times,the concent trend of Z-Ligustilide and E-Ligustilide was gradually increased from August to September,the highest in October and then decreased.The content trend of Butydenephthalide and Isobutydenephthalide was on the contrary. In the third chapter,it was studied on the dynamic accumulations of Ferulic acid, Ligustilide,Butydenephthalide of Angelica sinensis by UPLC-PDA in different regions and harvest times.It also conducted a cluster analysis and similarity evaluation.The content of results showed:Ligustilide>Ferulic acid>Butydenephthalide,the content trend of Ferulic acid and Ligustilide was gradually increased from August to September,the highest in October,but the content trend of Butydenephthalide was on the contrary.The result of the cluster analysis and similarity evaluation showed high similarity of Angelica sinensis in mature from different regions.The consistency was better,it can be clustered.In the third part,there were three chapters.It was about the quality standard assessment method of Angelica sinensis.In the first chapter,it was studied on TLC.The developer of TLC of Angelica sinensis of 2005 version in "Chinese Pharmacopoeia" was the toxic agent--benzene, in this chapter it added the TLC of Ligustilide and Butydenephthalide and changed the the toxic agent.This method was simple and reproducible.In the second chapter,It was studied on the content determination of volatile oil, polysaccharides and other main active ingredients.The method of content determination of volatile oil was established by pre-experiment.The average content was 0.30%~0.66%and we setted up the content limits.The method of content determination of polysaccharides was established.The average contents of netural and acidic polysaccharides were 11.25 mg/g~62.36 mg/g,19.02 mg/g~43.97 mg/g.The total polysaccharides was 11.25 mg/g~62.36 mg/g and we setted up the content limits.The method of simultaneous determination of Ferulic acid,Ligustilide, Butydenephthalide was established by UPLC-PDA and setted up the limits of content.In the third chapter,the method of fingerprint of Angelica sinensis was setting up by UPLC-PDA.The results of hierarehial cluster anaylsis and similarity evaluation showed that the shorter of storage time,the higher of similarity and convergence.It revealed there must be some relevance between aggregation types and similarity with the storage time.The main features and innovations in this dissertation are presented:1.Analyzed and evaluated multi-component targets(such as polysaccharides, volatile components,and ferulic acid) of Angelica sinensis and summed up the dynamics regular pattern of Angelica sinensis in different regions and harvest times. 2.Setted up a rapid analysis method of simultaneous determination of Ferulic acid,Ligustilide and Butydenephthalide by UPLC-PDA,setted up a fingerprint research methods of Angelica sinensis and setted up a TLC method by determination of Ferulic acid,Ligustilide and Butydenephthalide.It improved the quality control of Angelica sinensis further.
