Observation Of Curative Effect Of QingChangHuaShi,YiQiJianPi Method Sequential Treating Ulcerative Colitis And Experimental Study On Regulating PPAR-γ And NF-κB

Objective The aim of this study is to observe the clinical effect of QingChangHuaShi,YiQiJianPi method sequential treating ulcerative colitis;to observe the effect of this method on the expression of PPAR-γand NF-κB in colonic tissue of rats,and to explore the possible action mechanism through animal experiment.Methods Clinical part:Sixty patients who were diagnosed as in active stage of UC by colonoscopy and diagnosed as the syndrome of internal stasis of dampness and heat by syndrome differentiation of traditional Chinese medicine were divided into two groups,in treatment group,30 patients were treated with taking QingChangHuaShi prescription in combination with retention with enema prescription in activity period,then in remission period,the patients were treated with taking FuZhengQingChang prescription;while the other 30 patients in the control group were treated with particle release mesalazine(5-ASA).When the treatment is over, compare the clinical curative effect,the states of the intestinal mucous membrane and the remission rate.Experimental part:The SD rats were randomized divided into 4 groups as follows:normal control group,modell control group, QingChangHuaShi Presciption treatment group and Balsalazide treatment group. Make rat models of ulcerative colitis with TNBS and detect the expression PPAR-γand NF-κB in the rat's colon tissue by the method of RT-PCRResults Clinical research:The effect of improving symptoms of diarrhea,mucous bloody stool,abdominal pain,distension,tenesmus,fatigue in treatment group was better than that in control group(P＜0.05),but there was no statistical difference between the effect of improving anal burning and anorexia of the two groups(P＞0.05). There was statistical difference between the total effective rate of the two groups, and the curative effect in treatment group was better than that in control group (P＜0.05).The effect of improving the lesions of colonic mucosa in treatment group was better than that in control group(P＜0.05).The effect of improving the remission rate in treatment group was better than that in control group (P＜0.05).Experimental results:The expression of PPAR-γin colonic tissue in QingChangHuaShi prescription group was more than that in model group,while the expression of NF-κB in colonic tissue in QingChangHuaShi prescription group was less than that in model group(P＜0.05).Conclusion The curative effect of QingChangHuaShi,YiQiJianPi method Sequential treating ulcerative colitis patients is better than that of 5-ASA,especially in improving TCM syndrome,intestinal mucous membrane and remission rate.According to the experimental study,we conjecture that the mechanism of QingChangHuaShi prescription may be restraining inflammation and regulating immune through the expression of PPAR-γand NF-κB.
