Objective: To explore the interactions of Taohongsiwu Decoction's (THSWD's) effective constituents and the mechanism that they intervenes blood vessel regeneration and reconstruction of bone stump and consequently promotes the healing of fracture.Methods: In order to study on the interactions of Taohongsiwu Decoction's (THSWD's) effective constituent and Molecular Mechanism of it promotes the healing of fracture, Based on the former experiment,we extracted 4 effective constituents from THSWD:A(total alkaloids TA) who is the principal,B(total glycosides TG),C(total polysaccharide TPS) and D(volatile oil TVO) who is the secondary, executed experiment on the mus musculus albus after bare-handed fixed-point bone snaped, layed 8 groups according to the orthogonal design proposal. Moreover, the other groups of mus musculus albus (model group and origin group,9 and 10 group) reared at the same time was chosen as anormal contrast.Then we distributed randomly 120 mice to the above 10 groups according to the random digits table. At the 5th, 15th and 25st day after treatment,we took tissue of bone stump (callus) in every group above, analyzed blood vessel regeneration and observed the dynamic change of VEGF protein expression.Results:.In the histomorphology of callus, the quantity of neogenesis osteocytethe and the reforming process of callus in the1~8 group and 10 group were significantly more than that in the model group (P<0.01). In the micrangium morphosis of callus, the 5th, 15th day after treatment, micrangium morphosis of callus multiplied extensively, and the quantity of micrangium in the1~8 group and 10 group was significantly more than that in the model group (P<0.01). However, from the 21st day to the 35th day after treatment, the quantity of blood vessel of callus decreased gradually, and the morphosis trended to normal bone tissue, and regenerating blood vessel reached the reforming process, and the phase of reconstruction and remodeling of the vessels of callus tissue advanced more in 1~8 group and 10 group than in the model group. In VEGF protein of callus, VEGF expression quantity was more in 1~8 group and in 10 group than the model group(P<0.01), retard the decreasing trend of expression obviously in the later phase. The orthogonal experiment indicated that there was obvious difference between B's level 1 and level 2(P<0.05).But there was no difference between C's or D's level 1 and level 2(P>0.05).Conclusion: THSWD's effective constituents and their different compositions could improve the VEGF protein expression of callus tissue in bone stump. They could maintain higher expression environment in the region of bone stump where it stimulates proliferation of regenerated blood vessel of callus in early phase and accelerates the reconstruction of micrangium in callus in the later phase. They could accelerate blood swelling absorption and promote the growth and remodeling of the callus, then promote the healing of fracture.And there was interactions between TA and TC,but there was no interactions among TA,TPS and TVO,and so did among TC,TPS and TVO.
