| OBJECTIVE①To explore the essence of Liver Yang Forming Wind Syndrome(including Cerebral hemorrhage,Cerebral infarction,Cervical spondylosis,Parkinson disease) by Establishing the two dimensional polyacrylamide gel electrophoresis(2-DE) expression maps of total protein in PBMCs of the and the normal persons and analysing the different expression proteins.②To explore the essence of the Liver Yang Forming Wind Syndrome by comparing mRNA expression of PRX3 mRNA in the Liver Yang Forming Wind Syndrome and the normal persons.METHODS PBMCs of the Liver Yang Forming Wind Syndrome(including Cerebral hemorrhage,Cerebral infarction,Cervical spondylosis,Parkinson disease),and the normal persons were isolated by using Lymphozytes separation medium.2-DE was applied to separate the total proteins of the three groups to gain different epression proteins and estabilish 2-DE image and analyse them by PDQuest 7.3.1 software. Different epression protein were identified by MALDI-TOF-MS and database analysis.RT- PCR was used to determine the expressions of PRX3 in the two groups.RESULTS1.The expression maps of proteomics in PBMCs of Liver Yang Forming Wind Syndrome(including Cerebral hemorrhage,Cerebral infarction,Cervical spondylosis,Parkinson disease) and normal persons were establishied.Compared with the normal control group,Liver Yang Forming Wind Syndrome of Cerebral hemorrhage has 21 identical peotein spots,and has 17 different protein spots,and 13 proteins expression have down,4 proteins expression have up.Compared with the normal control group,Liver Yang Forming Wind Syndrome of Cerebral infarction has 22 identical peotein spots,and has 22 different protein spots,and 18 proteins expression have down,3 proteins expression have up.Compared with the normal control group,Liver Yang Forming Wind Syndrome of Parkinson disease has 23 identical peotein spots,and has 20 different protein spots,and 18 proteins expression have down,2 proteins expression have up.Compared with the normal control group,Liver Yang Forming Wind Syndrome of Cervical spondylosis has17 identical peotein spots,and has 15 different protein spots,and 13 proteins expression have down,2 proteins expression have up.And have 6 same proteins expression in Liver Yang Forming Wind Syndrome.2.Compared with the normal control group,Thioredoxin-dependent peroxide reductase decreased in the liver-yang forming wind group(p<0.05).CONCLUSION1.The expression maps of proteomics in PBMC_s of with Liver Yang Forming Wind Syndrome(including Cerebral hemorrhage,Cerebral infarction,Cervical spondylosis,Parkinson disease) and normal persons were establishied.2.The Liver Yang Forming Wind Syndrome have identical proteins and different proteins,which means that different diseases with the card with the same material foundation.3.Thioredoxin-dependent peroxide reductase decreased in the liver -yang forming wind group,indicating that the Syndromes have same substance basis.4.The Liver Yang Forming Wind Syndrome of different diseases with the same card has expression of PRX3,in every groups of the liver -yang forming wind group(including Cerebral hemorrhage,Cerbral infarction Cervical spondylosis,Parkinson disease) PRX3 expression of no difference,PRX3 may become of the specific indicators of liver - yang forming wind. |
