OBJECTIVEâ‘ To explore the essence of the Liver Yang Syndrome (HOLYS) and Liver Yang Forming Wind Syndrome (LYFWS) by Establishing the two dimensional polyacrylamide gel electrophoresis (2-DE) expression maps of total protein in PBMC_S of the the primary hypertension patients with HOLYS, the hypertension hemorrhage patients with LYFWS and the normal persons and analysing the different expression proteins.â‘¡To explore the essence of the HOLYS and LYFWS by comparing mRNA expression of GSTO1 mRNA in the HOLYS, LYFWS and the normal persons.METHODS PBMC_S of the hypertension intracerebral hemorrhage patients with Liver Yang Forming Wind Syndrome(LYFWS), the primary hypertension patients with Hyperactivity Of Liver Yang Syndrome (HOLYS), and the normal persons were isolated by using Lymphozytes separation medium. 2-DE was applied to separate the total proteins of the three groups to gain different epression proteins and estabilish 2-DE image and analyse them by PDQuest 7.3.1 software. Different epression protein were identified by MALDI-TOF-MS and database analysis.RT-PCR was used to determine the expressions of GSTO1 in the three groups. RESULTS1. The expression maps of proteomics in PBMC_S of primary hypertension patients with HOLYS, hypertension intracerebral hemorrhage patients with LYFWS, and normal persons were establishied . 18 different protein spots and 6 identical peotein spots were found in HOLYS group and LYHF group.The identified proteins include cytoskeletal proteins, metabolic enzymes,signal transmission proteins ,antioxydation proteins blood proteins and so on.2. Compared with the normal control group, Glutathione transferase omega-1 iecreased in the the liver -yang hyperactivity group and in the liver-yang forming wind group(p<0.05). But the Glutathione transferase omega-1 increased significantly in the liver -yang hyperactivity group than in the liver - yang forming wind group(p<0.05).CONCLUSION1. The expression maps of proteomics in PBMC_S of primary hypertension patients with HOLYS, hypertension intracerebral hemorrhage patients with LYFWS, and normal persons were establishied.2. The HOLYS and LYHFS of primary hypertension have identical proteins and different proteins,which means that The HOLYS and LYHFS may have same matter foundation and have different essence .3. Glutathione transferase omega-1 increased in the liver -yang hyperactivity group and in the liver - yang forming wind group, indicating that the tow Syndromes have same substance basis . But it increased significantly in the liver -yang hyperactivity group than in the liver - yang forming wind group. It indicates that The HOLYS and LYHFS may have different essence. This may lay a foundation for the further investigasion of essence of the LYFWS.
