Objectives:Diabetes mellitus is a common and frequently occurring disease.It has become increasingly clear that both genetic and environmental factors contribute to the etiology of diabetes mellitus (DM).Discovering ang identify the predisposing genes is very important to finding the population at high risk for diabetes mellitus.A new apolipoprotein gene,ApolipoproteinA5(APOA5),was recently discovered near the APOA1/C3/A4 gene cluste.Many studies in different ethnic populations have demonstrated associations of some of the APOA5 SNPs with plasma TG.Recently the APOA5-1131T/C was paid attention.Thus,our objective is to investigate the relationship between APOA5 gene-1131T/C polymorphism and type 2 diabetes mellitus in Han nationality.Methods:Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) were used to analyze the APOA5-1131T/C polymorphism in 484 patients with T2DM and 538 controls.The levels of serum lipids profiles were also measured by enzymatic methods.Results:1.TheAPOA5-131T/C genotype distribution in T2DM group and the control group showed a significant deference(P<0.05).Compared with the control group,type CC was higher and typeTT was lower in T2DM group.The frequency of the-1131C allele in patients with T2DM was significantly higher than that of the controls(0.324vs.0.258,P<0.05).2.The serum TG levels in three genotype groups showed a significant diference(P=0.01).The TG levels in typeTC(1.68±1.82mmol/L) and typeCC(2.02±2.32mmol/L) were significantly higher than in typeTT(1.44±1.32mmol/L)(all P<0.01),and the HDL-C levels in the three types was also different.In controls the TG levels was also different among the three genotype(0.94±0.33 mmol/L vs.1.09±0.62 mmol/L vs.1.01±0.30 mmol/L,P=0.026).Compared with the noncarriers,the C carriers also had a higher TG and HDL-C levels in T2DM group(1.08±0.59mmol/Lvs.0.94±0.33mmol/L and 1.40±0.27mmol/L vs.1.41±0.23mmol/L,all P<0.05),but the levels of TC and LDL-C did not show statistical differences in various genotypes(P>0.05).3.Unadjusted logistic regression analysis indicaded that TC+CC genotype of APOA5-1131T/C had a increased T2DM risk(OR=1.546, 95%Cl 1.207-1.979,P=0.001).And the analysis adjusted for age,sex,BMI,SBP,FBS,TG and HDL-C also show statistical differences (OR=1.339,95%CI 1003-1.778,P=0.048).Conclusions:The frequency of theAPOA5-1131C allele in patients with T2DM was significantly higher than that of the controls.And that allele was associated with T2DM susceptibility.APOA5-1131T/C polymorphism has a significant influence on human serum TG and HDL-C levels and the APOA5-1131T/C variant is significantly associated with increased serum TG levels and decreased serum HDL-C levels.
