| Objective: The purpose of this study was to screen out the related risk factors of Mongolian people with high triglyceride combined with type 2 diabetes in Eastern Inner Mongolia,and explore the relationship between the single nucleotide polymorphism(SNP)site rs7578326 of insulin receptor substrate-1(IRS1)gene and Mongolian people with high triglyceride combined with type 2 diabetes,and analyze the relationship between different genotypes and the genetic susceptibility of Mongolian people with high triglyceride combined with type 2 diabetes.To explore the possible risk factors of Mongolian people with high triglyceride combined with type 2 diabetes at the gene level,and provide scientific basis for intervention and prevention policies of related diseases.Methods: Using molecular biological research methods such as questionnaire survey,detection of physiological and biochemical indexes and physical examination,Polymerase chain reaction,second generation sequencing,Sanger sequencing and taking the rs7578326 polymorphism of IRS1 gene as the target.SPSS statistical method was used to analyze and determine the genotype and allele frequency differences between different groups and the possible risk factors of Mongolian high triglyceride combined with type 2 diabetes.Results :1、In this study,the subjects were divided into normal control group(NC group),type 2diabetes group(T2DM group)high triglyceride combined with type 2 diabetes group(High TG+T2DM group)and high triglyceride group(High TG group).Statistical analysis of the general condition of the study subjects between groups showed significant differences in Age,BMI,SBP,DBP,WC,TC,TG and LDL-C levels(p<0.05).2、Logistic stepwise regression analysis showed that in the history of diabetic disease,TG levels,TC levels and increase of BMI index may increase the risk of T2 DM combined with high triglycerides in Mongolians(OR>1,p<0.05).Mongolians with elevated HDL cholesterol levels had a reduced risk of high triglyceride combined with T2DM(OR<1,p<0.05).3、The allele and genotype distribution at the rs7578326 locus of the IRS1 gene conform to the Hardy-Weinberg law of genetic equilibrium.After adjusting for confounders such as BMI and LDL-C,an association was found between the(AG+GG)genotype and Mongolian hypertriglyceridemia combined with type 2 diabetes in the dominant model(p<0.05).The rs7578326 locus was not found to be associated with Mongolian hypertriglyceridemia combined with type 2 diabetes in either a co-dominant or recessive genetic model.4、In the Mongolian high triglyceride combined with type 2 diabetes group,TG levels and LDL-C levels in(AA+AG)group were significantly higher than those in GG group(p<0.05).Conclusions: The disease history of diabetes,TG level,TC level and BMI index may be the risk factors of Mongolian people with high triglyceride combined with type 2 diabetes.The IRS1 gene SNP rs7578326 polymorphism is associated with the susceptibility of Mongolian people with high triglyceride combined with type 2 diabetes.Allele A at SNP rs7578326 of IRS1 gene is associated with Mongolian people with high triglyceride combined with type 2 diabetes. |
