Objective:To observe the changes of heat shock protein70(HSP70) expression in peripheral white blood cells and serum on severely burned patients.To investigate the effect role of Intracellular and Extracellular Heat Shock Protein70 on severely burned patients.Methods:This experiment was divided into 3 groups:the normal resuscitation group on post burn day(PBD), the delayed resuscitation group on PBD,and the control group. There are 10 cases in each group.Expression of HSP70 in peripheral white blood cells was detected by reverse transcription polymerase chain reaction (RT-PCR) . The level of serum HSP70 was measured by enzyme-linked immunosorbent assay(ELISA).Results:①RT-PCR result showed that:The expressive intensity of HSP70 on 3 PBD of the normal resuscitation group(1.2351±0.3439 ) was remarkably higher than that on 1 PBD (0.8711±0.1751),21 PBD (0.3978±0.1139)and the control group (0.3287±0.1047), There was a significant statistical significance (P <0.05);The expressive intensity of HSP70 on 1 PBD was remarkably higher than that 21 PBD and the control group, (P <0.05);The expressive intensity of HSP70 on 1 PBD (1.2689±0.2775) and 3 PBD (1.3832±0.3001) of the delayed resuscitation group was remarkably higher than that on 21 PBD (0.4283±0.1222)and the control group (0.3287±0.1047),(P <0.05);But the expressive intensity of HSP70 on 3 PBD was a little higher than that on 1 PBD,there was not a statistical significance (P >0.05).The expressive intensity on 21 PBD in the normal resuscitation group,the delayed resuscitation group and the control group was not a statistical significance (P >0.05) ;But the expressive intensity of HSP70 on 1 PBD of the delayed resuscitation group was remarkably higher than that on 1 PBD of the normal resuscitation group.②ELISA result showed that:The level of serum HSP70 of early state in the normal resuscitation group and the delayed resuscitation group had remarkably increased,the peak time in the normal resuscitation group was 3 PBD,but the peak time in the delayed resuscitation group had significantly retruded 7 PBD,and then the level of serum HSP70 in two group had gradually decreased to near the normal levels.Conclusions:The expression of Intracellular and Extracellular HSP70 could be detected HSP70 after burns.Severe burns could induced the enhanced expression of HSP70,and the changed expression of HSP70 was associated with development of burns.The expression and change of HSP70 may play some role in onset and progressive of burns,HSP70 may protect organism from damage and promote the healing .It is of great clinical value in diagnosing the severity of burns.
