Objective:To investigate the changes of HSP60 and HSP90 in patients with gout arthritis(GA)and their possible roles.Methods:(1)a total of 158 male gout patients(GA),including 79 patients with acute gout(Ag)and 79 patients with intermittent gout(Ig),were collected from January 2020 to August 2020 in Nanchong central hospital.One hundred and twenty healthy controls(HC)who underwent physical examination in the physical examination center of Nanchong Central Hospital during the same period were selected as the control group.The clinical data,laboratory indexes and peripheral venous blood of the experimental group and the control group were collected.The serum was collected and the mononuclear cells(PBMCs)were isolated.The RNA was extracted and the cDNA was reversed.(2)Enzyme linked immunosorbent assay(ELISA)was used to detect the serum levels of HSP60 and HSP90 in each group.(3)Real time quantitative polymerase chain reaction(RT qPCR)was used to detect the expression of HSP60 and HSP90 mRNA in PBMCs.(4)Objective to analyze the correlation between the expression levels of HSP60 and HSP90 in serum of patients with gout and their clinical and laboratory indexes.(5)SPSS 17.0 statistical software was used for analysis.Wilcoxon rank sum test and one-way ANOVA were used for comparison among measurement data groups.Spearman correlation analysis was used for correlation between variables.Results:(1)the serum levels of HSP60 and HSP90 in GA group were significantly higher than those in HC group(7.5±0.4pg/ml vs 1.5± 0.8;7.3± 0.2 vs 1.5 ± 0.07,all P<0.05),and those in Ag group were higher than those in Ig group(9.8± 0.6 vs 5.2 ± 0.1;9.2± 0.1 vs 5.3± 0.1,all P<0.05).(2)In PBMCs,the expression levels of HSP60 and HSP90 in HC group(219± 5.8216± 5.8)were significantly lower than those in GA group(638 ±16743± 40),and those in Ag group(840± 11851± 11.4)were significantly higher than those in Ig group(521±14631± 78)(all P<0.05).(3)Correlation analysis showed that HSP60 level in serum and PBMCs was positively correlated with PLT,CRP and ESR(r=0.238,P=0.02;r=0.597,P=0.01;r=0.597,P=0.01);HSP90 level was positively correlated with WBC,PLT,CRP and ESR(r=0.184,P=0.021;r=0.244,P=0.002;r=0.660,P=0.000;r=0.744,P=0.000);HSP60 mRNA was positively correlated with PLT,CRP and ESR(r=0.191,P=0.016;r=0.537,P=0.000;r=0.708,P=0.000),HSP90 mRNA was positively correlated with PLT,CRP and ESR(r=0.191,P=0.017;r=0.546,P=0.000;r=0.693,P=0.000).Conclusion:the expression of HSP60 and HSP90 in peripheral blood of patients with gout is significantly increased,suggesting that HSP60 and HSP90 may be involved in the occurrence of gouty arthritis.
