This thesis includes the following three chapters:Chapter 1 The research progress were introduced about Radix et Rhizoma Rhei,Rhizoma Coptidis,Radix Scutellariae and its preparations. It is narrated that the purpose and the main contents of this study.Chapter 2 To establish a method for the determination of Wogonoside;Wogonin;emodin; aloe-emodin; rehein; chrysophanol; physcion in Sanhuang Pian and Yiqing Jiaonang and Yiqing Keli by micellar electrokinetic capillary chromatography. The separation was performed on a uncoated fused silica capillary of 55cm×75μm ID. (47cm effective length)25mmol·L-1Na2B4O7buffer+25mmol·L-1SDS+4mmol·L-1SBE-β-CD+8%Methanol solution(pH9.42)was selected for the running buffer. The voltage applied was 14kV. The sample was injected by gravity(5s,15cm). The detection wavelength was 254 nm. The internal standard was acetaminophen.The linear ranges of determination for Wogonoside;Wogonin; emodin;aloe- emodin; rehein;chrysophanol;physcion in Sanhuang Pian were 2.02~20.16μg·ml-1(r = 0.9996),1.0~10.0μg·ml-1(r = 0.9993),1.6~16.0μg·ml-1(r = 0.9996),1.2~12.0μg·ml-1(r = 0.9990),2.0~20μg·ml?(1r = 0.9995),7.2~72 .0μg·ml?(1r = 0.9986),1.0~10.0μg·ml-1(r =0.9981)separately. The average recovery of Wogonoside was 100.6%, precision of the method was 1.15%(RSD, n=6). The average recovery of Wogonin was 98.3%, precision of the method was1.51% (RSD, n=6). The average recovery of emodin was 99.9%, precision of the method was 2.81% (RSD, n=6). The average recovery of aloe-emodin was 101.2%, precision of the method was 1.49%(RSD, n=6). The average recovery of rehein was 99.8%, precision of the method was 2.77%(RSD, n=6). The average recovery of chrysophanol was 102.0%, precision of the method was 2.60%(RSD, n=6). The average recovery of physcion was 101.8%, precision of the method was 2.04% ( RSD, n=6).The linear ranges of determination for Wogonoside;Wogonin; emodin;aloe- emodin; rehein;chrysophanol;physcion in Yiqing Jiaonang were 2.02~20.16μg·ml-1(r = 0.9996),1.0~10.0μg·ml-1(r = 0.9993),1.6~16.0μg·ml-1(r = 0.9996),1.2~12.0μg·ml-1(r = 0.9990),2.0~20μg·ml-1(r = 0.9995),0.8~8.0μg·ml-1(r = 0.9997),0.2~2.0μg·ml-1(r = 0.9996). The average recovery of Wogonoside was 102.0%, precision of the method was 0.65% (RSD, n=6). The average recovery of Wogonin was 98.8%, precision of the method was3.02% (RSD, n=6). The average recovery of emodin was 99.5%, precision of the method was 2.87% (RSD, n=6). The average recovery of aloe-emodin was 101.2%, precision of the method was 3.12% (RSD, n=6). The average recovery of rehein was 100.4%, precision of the method was 1.69% (RSD, n=6). The average recovery of chrysophanol was 99.3%, precision of the method was 2.32% (RSD, n=6). The average recovery of physcion was 99.5%, precision of the method was 2.01% (RSD, n=6).The linear ranges of determination for Wogonoside;Wogonin; emodin;aloe- emodin;chrysophanol;physcion in Yiqing Keli were 2.02~20.16μg·ml-1(r = 0.9996),1.0~10.0μg·ml-1(r = 0.9993),1.6~16.0μg·ml-1(r = 0.9996),1.2~12.0μg·ml-1(r = 0.9990),7.2~72 .0μg·ml-1(r = 0.9986),1.0~10.0μg·ml-1(r = 0.9981). The average recovery of Wogonoside was 102.7%, precision of the method was 2.16% (RSD, n=6). The average recovery of Wogonin was 101.8%, precision of the method was 2.61% (RSD, n=6). The average recovery of emodin was 102.0%, precision of the method was 2.22% (RSD, n=6). The average recovery of aloe-emodin was 99.0%, precision of the method was 1.82% (RSD, n=6). The average recovery of chrysophanol was 100.0%, precision of the method was 1.71% (RSD, n=6). The average recovery of physcion was 98.2%, precision of the method was 3.06% (RSD, n=6).The method proved to be convenient,rapid and well repeatable,and can be used for quantitative control of Sanhuang Pian and Yiqing Jiaonang and Yiqing Keli. Chapter 3To establish a method for the determination of Berberine hydrochloride in Yiqing Keli and Yiqing Jiaonang by capillary zone electrophoresis.The separation was performed in a uncoated fused silica capillary of 60cm×75μm ID (52cm effective length). 0.2mol·L-1 sodium dihydrogen phosphate+ethanol(1:1)(pH5.50)was selected for the running buffer. The voltage applied: was 21kV, The sample was injected by gravity(10s,15cm). The detection wavelength was 265 nm. The internal standard was ranitidine hydrochloride. The linear ranges of determination for Berberine hydrochloride was 2.613.0μg·ml-1 (r= 0.9972), The average recovery of Berberine hydrochloride in Yiqing Keli was 100.3%, precision of the method was 0.96% (RSD, n=6). The average recovery of Berberine hydrochloride in Yiqing Jiaonang was 99.4%, precision of the method was 1.86% (RSD, n=6).To establish a method for the determination of Palmatine hydrochloride and Jatrorrhizine hydrochloride in Yiqing Keli and Yiqing Jiaonang by capillary zone electrophoresis. The separation was performed in a uncoated fused silica capillary of 60cm×75μm ID (52cm effective length). 0.2mol/L sodium dihydrogen phosphate+ethanol(1∶1)(pH5.50)was selected for the running buffer. The voltage applied: was 18kV. The sample was injected by gravity(10s,15cm). The detection wavelength was 265 nm. The internal standard was ranitidine hydrochloride. The linear ranges of determination for Palmatine hydrochloride and Jatrorrhizine hydrochloride were 2.37~11.84μg·ml-1(r = 0.9991),0.448~2.24μg·ml-1(r = 0.9985). The average recovery of Palmatine hydrochloride in Yiqing Keli was 102.1%, precision of the method was 1.02% (RSD, n=6). The average recovery of Jatrorrhizine hydrochloride in Yiqing Keli was 98.5%, precision of the method was 1.29% (RSD, n=6). The average recovery of Palmatine hydrochloride in Yiqing Jiaonang was 99.3%, precision of the method was 2.58% (RSD, n=6). The average recovery of Jatrorrhizine hydrochloride in Yiqing Jiaonang was 99.7%, precision of the method was 2.75% (RSD, n=6).The method proved to be convenient,rapid and well repeatable,and can be used for quantitative determination of Yiqing Keli and Yiqing Jiaonang.
