Traditional Chinese medicine is a multi-component complex system of varietychemical compositions, identifying the ingredients by comparing chemical referencesubstance is difficult, so regarding the component peaks as a whole, which can translateintrinsic characteristics into drugs fingerprint and we call it TCM fingerprint technology.Information reflect or obtain by the TCM fingerprint is so abundance and complex that weneed to process and analysis the original data by intuitive comparison, quantitative dataanalysis, cluster analysis, principal component analysis or artificial neural network analysis,etc. Curve matching method has a wide application in the international arena, such as thestudy of the growth curve, the elderly gait research, hormonal time series studies, geneexpression, image recognition, but it has never been used on the TCM fingerprint analysis.This thesis is including three parts. Firstly, to summarize the fingerprinting researchprogress. Second, to nanlyze curve matching method applied to the fingerprints of singleherb rhubarb, compound and compound serum. Third, to study the active ingredient contentdetermination.In the first section, commonly used research methods and technology of fingerprint,evaluation methods in TCM fingerprint, similarity evaluation methods and the applicationof evaluation method has been summarized.In the second part, take rhubarb, Yiqing soft capsule which contain rhubarb and it’sserum for example, analysis curve matching method used in fingerprint data processing.First of all, to establish UPLC chromatographic fingerprints of14different originrhubarb samples and three standard reference drug, conduct peak selection, spectra alignment and comparison by curve matching method, we can find8common chemicalcompositions in rhubarb which comes from different origin, then using the substancereference and HPLC-MS method we can identify the8common components, they aremono-galbyl-3-O-glucose, mono-galbyl-4-O-glucose, gallic acid,physcion-8-O-β-D-monoglucoside, aloe-emodin, emodin, chrysophanol and physcionrespectively. This study provides a new idea and new way for common medicinalcomponents analysis of the same kind Chinese herbal medicines from different origin.Besides, similarity evaluation, cluster analysis and principal component analysis arecompared, and the results show that the similarity of rhubarb from different origin is low,so the similarity analysis has its limitations; the clustering analysis and similarity analysisshows the same result that cannot judge the quality of rhubarb or distinguish commerciallyrhubarb; principal component analysis point out4component which is more important thanother15common ingredients, by comparing the content of these4components, combinewith cluster analysis results and the view of overall spectrum, we can determine theoptimum sample.What’s more, to further study of the curve matching method apply in compoundfingerprint, fingerprint of five manufacturers of15batches of Yiqing soft capsule areestablished,14common components are obtained by curve matching method analysis, andthrough substance reference4common components are identified, they are berberinehydrochloride, baicalin, rhein and emodin. Compare similarity analysis, cluster analysis andprincipal component analysis method applying to Yiqing soft capsule’s quality evaluation,the result shows that cluster analysis and similarity analysis can be used to distinguishYiqing soft capsule sample from different manufacturers, and principal component analysispoint out7more important components, and four of them are the common ingredients findby curve matching analysis. Through the comparison of reference and single herb,attributable the characteristic peaks.Finally, to study the material basis of Yiqing soft capsule, administer rats with twomanufacturers’ Yiqing soft capsule, and the serum transitional components are detected, theamount are18,23respectively, and emodin is the prototype component transfer into the blood.The third section, to establish a method for simultaneous quantitative determination of8active components in Yiqing soft capsule, the results show that the content of emodin andbaicalein is high, and the8index components’ content is stable between different batches.Adopt orthogonal test combinative with comprehensive multiple index evaluation toexplore the best extractive craft of samples.The innovation in this paper: The commonly used similarity analysis methods intraditional medicine fingerprint still need to select peak by man first, its greater subjectivity.Curve matching method has never been used in fingerprinting. The method apply in TCMfingerprint data processing, through a series of system analysis method rather thandescriptive, combine all the features of the chromatogram information to achieve moreaccurate fingerprint judgment, and summing up a standard fingerprint by large amounts ofdata, which can be used to look for the common chemical compositions directly in differentorigin medicine or compound preparation from different manufacturers.
