Objective:This topic take the Basic Theory of Traditional Chinese Medicine as the instruction, the purpose was to observe the treatment effects of the Yu Siling 1# Fang on changing the ovary morphology and E2, P, T of the blood serum of the female rats anovulatory due to testosterone model,and revelating the action mechanism of the Yu Siling 1# Fang on treating Anovulatory Infertility, providing the scientific basis for the validity of he Yu Siling 1# Fang on treating Anovulatory Infertility.Methods:The anovulation rats model was established by injecting testosterone propionate subcutaneously to SD female rats of 9 days age.By using smear of the rats vaginal exfoliated cells to examine the model whether or not succeed. Successful model of the rat entered the next experiment, after 28-day's gavaging , to observe the weight of the ovarian of each group, to observe the ovarian morphological changes of each group by light microscope, Detected the E2, P, T levels in each group serum by RIA.Results:1 Effect on rat-ovarian-weight coefficientThe rat-ovarian-weight coefficients of the normal group, model group and treatm -ent group( x±s)were 0.3766±0.0522,0.3009±0.0452 and 0.3622±0.0402. The rat- ovarian-weightcoefficient of the model group was smaller than the normal group's ,the difference was significant(P<0.01), the treatment group's and thenormal group's was nearly ,there was no significant difference(P>0.05); There was significant difference during the the treatment group's and the normal group's(P <0.01).2 Effect on ovarian morphology of rat2.1 The general observation of ovarianThe ovarian color of the 10 rats of the normal group was bright red. The ovarian color of the 10 rats of the model group was pale. The ovarian color of the 10 rats of the treatment group was bright red.2.2 Examination of ovarian by optical microscopicThere were a large number of luteums and follicles of all developmental stages in the rats'ovarians of the normal group, the granulosa cell layers were thick (6-13 layers), arranged neatly; There were cystic follicles in the model rats'ovarians by the microscope, the granulosa cell layers were thin (1-2 layers), arranged loosely, three of ten model rats'ovarys had a small amount of ovarian luteums, but the quantity was smaller than the normal group's ,the other seven rats had no ovarian luteum; The structure of rat's ovarian of the treatment group was improved , there were cystic expansion of the follicles,the development follicles, and a large number of luteums and well-developed ovarian follicles at all levels , the granulosa cell layers were thick (4-8 layers), arranged neatly.3 Effect on E2,P,T levels in each group rat's serum3.1 Serum E2 comparisonThe serum E2 of the normal group, model group and treatment group( x±s) were 18.12±2.00,13.09±1.89 and 17.93±2.84. The serum E2 of the model group was smaller than the normal group's ,the difference was significant(P<0.01), the treatment group's and thenormal group's was nearly ,there was no significant difference(P>0.05); There was significant difference during the the treatment group's and the normal group's(P <0.01).3.2 Serum P comparisonThe serum P of the normal group, model group and treatment group( x±s) were 10.85±2.13,9.00±1.72 and 10.87±1.34. The serum P of the model group was smaller than the normal group's ,the difference was significant(P<0.05), the treatment group's and thenormal group's was nearly ,there was no significant difference(P>0.05); There was significant difference during the the treatment group's and the normal group's(P <0.05).3.3 Serum T comparisonThe serum T of the normal group, model group and treatment group( x±s) were 32.24±1.92,63.26±1.99 and 32.81±2.05. The serum T of the model group was smaller than the normal group's ,the difference was significant(P<0.01), the treatment group's and thenormal group's was nearly ,there was no significant difference(P>0.05); There was significant difference during the the treatment group's and the normal group's(P <0.01).Conclusions:1 The Yu Siling 1# Fang can be significantly increased the rat-ovarian-weight coefficient of the anovulatory rat.2 The Yu Siling 1# Fang can be significantly improved the structure of ovarian of the anovulatory rat,was good for the follicular development, increased the number of ovarian follicles at all levels and the number of luteums;3 The Yu Siling 1# Fang can significantly improve the levels of the serum E2,P,T of the anovulation rat, return the imbalanced E2,P,T to normal.
