Objective To study the stroke of stroke-prone Renovascular Hypertensive Rats subjected artificial cold and changes of serum von Willebrand factor (vWF) and interleukin-6 (IL-6) in stroke-prone Renovascular Hypertensive Rats before subjected artificial cold. To explore how artificial cold effect the stroke of RHRSP and how changes of serum vWF and IL-6 forecast stroke.Methods 250 male Sprague-Dawley rats were randomly divided into two groups: the hypertensive group and the sham-operated group. 200 rats were selected to make the 2-kidney, 2 clip (2k2c) renovascular hypertensive model. SBP was measured by an indirect tail-cuff sphygmomanometer (HX-1), in preheated (37°C, 15 minutes) conscious rats before renal artery constriction and every four weeks intervals after renal artery constriction for 12 weeks. two groups were put into the artificial climate cabinet. The artificial cold exposure was designed as 3 cycles of 12 h light of 22℃and 12 h dark of 4℃.Then all surviving rats were killed,and observed hematoxylin-eosin stained brain sections by routine light microscopic. the hypertensive group were divided into three groups: no-stroke group, cerebral infarction group and cerebral hemorrhage group by the result of pathology.Vein blood 1.5ml was collected from sublingual vein before artificial climate cabinet. vWF and IL-6 were measured with enzyme linked immunoabsorbent assay( ELISA). Results Results of stroke :There wasn't stroke in the sham-operated group after artificial cold and there were 11 cerebral infarction rats and 36 cerebral hemorrhage rats in the hypertensive group.The rate of stroke was 24.7%. The mean systolic BP in different stages :The mean SBP were112±12 mm Hg ,189±20 mm Hg ,192±19 mm Hg,213±21 mm Hg before subjected artificial cold in the sham-operated group, no-stroke group, cerebral infarction group and cerebral hemorrhage group. The mean SBP in the three hypertensive groups was significantly higher than that of the sham-operated group(P<0.001) ;The mean SBP in the cerebral infarction group and cerebral hemorrhage group were significantly higher than that of the no-stroke group(P<0.01) ;but there was no significant difference between the cerebral infarction group cerebral hemorrhage group( P > 0. 05). Plasma vWF levels in different stages :The plasma vWF levels were187.6±21.9 pg/ml ,225.3±20.1 pg/ml,190.1±24.8 pg/ml before subjected artificial cold in the no-stroke group, cerebral infarction group and cerebral hemorrhage group. The plasma vWF levels before subjected artificial cold was 100.5±23.2 pg/ml in the sham-operated group . So the plasma vWF levels in the no-stroke group, cerebral infarction group and cerebral hemorrhage group were significantly higher than that in the sham-operated group (P<0.001, P<0.01,P<0.01, respectively).There was significant difference of plasma vWF between the no-stroke group and cerebral infarction group(P<0.01) and between the no-stroke group and cerebral hemorrhage group(P<0.01) ,but there was not significant difference between the cerebral infarction group and cerebral hemorrhage group( P > 0. 05). Plasma IL-6 levels in different stages :The plasma IL-6 levels were174.5±27.5 ,245.4±22.7 ,230.8±24.2before subjected artificial cold in the no-stroke group, cerebral infarction group and cerebral hemorrhage group. The plasma IL-6 levels before subjected artificial cold was 115.9±28.5in the sham-operated group. So the plasma IL-6 levels in the no-stroke group, cerebral infarction group and cerebral hemorrhage group were significantly higher than that in the sham-operated group (P<0.001, P<0.01,P<0.001, respectively). There was significant difference of plasma IL-6 between the no-stroke group and cerebral infarction group(P<0.001) and between the no-stroke group and cerebral hemorrhage group(P<0.001) ,but there was not significant difference between the cerebral infarction group and cerebral hemorrhage group( P > 0. 05). Correlative analysis: Through correlative analysis, there is clear positive correlation between SBP and vWF (r=0.728, P < 0. 001) ,SBP and IL-6(r=0.745, P < 0. 001) , vWF and IL-6 (r=0.821, P < 0. 001).Conclusion 1. Serum von Willebrand factor (vWF) and interleukin-6 (IL-6) in stroke-prone Renovascular Hypertensive Rats were increasing.2. vWF and IL-6 significant increased before RHRSP have stroken subjected artificial cold,so vWF and IL-6 could Play an important role before RHRSP have stroken subjected artificial cold.
