Objective:Isolation and identification of 1 mononuclear cells subsets of BCR/ABL positive,FIK1~+,CD34~+,CD38~-expressed in bone marrow of patients with chronic myeloid leukemia,establish chronic myeloid leukemia NOD/SCID model in vitro,and preliminarily explore the biological properties of such cellsMethods:Extract 5ml bone marrow under sterile conditions from patients with chronic myeloid leukemia with positive BCR/ABL fusion gene,anticoagulant with heparin, separation of mononuclear cells with lymphocyte separation medium,then cultured in vitro,sorting FIK1~+,CD34~+,CD38~- cells using flow cytometry,injected the cells into the NOD/SCID mice according to density of 1×10~3,establish chronic myeloid leukemia nude model in vitro,extract bone marrow under sterile conditions from the nude mice model,anticoagulant with heparin,separation of mononuclear cells with lymphocyte separation medium,then identify its immune phenotype using flow cytometry analysis, detect characteristics of stem cells induced differentiation in vitro,and detected the expression of BCR/ABL fusion gene.Results:1.BCR/ABL positive,FIK1~+,CD34~+,CD38~- cells can differentiate into leukemia cells in receptor mice,and it can reconstruct similar disease as human chronic myeloid leukemia in receptor mice.2.BCR/ABL positive,FIK1~+,CD34~+,CD38~- cells have characteristics of stem cell;amplified with single clone in vitro and differentiate to hematopoietic cell. Conclusions:BCR/ABL positive,FIK1~+,CD34~+,CD38~- cells may be stem cells of chronic myeloid leukemia.
