Abstract/Summary:
Objective: To investigate and analyze the the efficacy and safety of the second generation tyrosine kinase inhibitors(dasatinib and nilotinib) for chronic myeloid leukemia patients in the course of treatment appeared resistant or intolerant.Methods:Observation the efficacy and safety of 11 patients with CML-CP patients and 5 cases CML-AP patients after imatinib failure or intolerance to switch 2nd TKI treatment in February 2009 to 2015 by the end of April.Results:11cases of CML patients obtained a CHR(93.75%), as the median duration of theraphy was 2(1-3)months,14 cases patients achieved MCyR(87.5%), 10 patients achieved CCyR(62.5%),7 cases received MMR(43.75%). After the second generation tyrosine kinase inhibitors in the treatmentof common hematologic adverse events included granulocyte reduction(18.75%), anemia(12.5%), thrombocytopenia(6.25%).Nonhematologic adverse events nausea, vomiting(25%), constipation(12.5%), arrhythmia(12.5%), in addition to rare oral ulcer, elevated transaminases, and rash.CML-AP happened grade 3-4 hematologic events,was more higher thanCP patients.Conclusion: The second generation tyrosine kinase inhibitors for resistant or intolerant patients of imatinib has good safety, tolerance, and as early as possible and regular and consistent application of drugs can enable patients to benefit more, also for patients waiting for better treatment time and opportunity.
