Objective To explore the effect and related mechanism of celery on reproductive system in male mice,providing evidence for further development and use of celery. Methods 240 healthy adult Kunming male mice of SPF were randomly divided into two time groups,which were seven-day and 56-day group.Then each time group were divided into four groups according to body weight,including negative control group,low-dose group,medium-dose group and high-dose group.Mice of each group were administered by gavage at 8:30am every day,control group with 0.3 milliliters of normal saline and others groups with 0.3 milliliters of celery juice of different concentration.Ten mice were selected randomly and killed at the end of intragastric administration,one and two weeks of recovery in each time group.(ⅰ) Liver,spleen, kidney,testis and epididymis were taken to be weighed and calculated organic indexes respectively.(ⅱ) The content of hemoglobin,ALT,AST,ALT/AST,ALB,GLO,A/G, ALP,the level of T,LH and FSH in serum was measured.(ⅲ) The activities of SOD, GSH-Px,MDA and T-AOC were measured.(ⅳ) Pathological alterations of testis in male mice were observed with lightmicroscope and ultramicrostructure with electronmicroscope.(ⅴ) Flow cytometry(FCM) was made to detect all kinds of ploidy cells of testicular spermatogenic cells and its cycle.Results(ⅰ) There were statistical significances in liver indexes between the medium-dose group and negative control group or high-dose group and negative control group(P<0.05) and there were no statistical significances in all kinds of organ indexes among all groups after one and two weeks of recovery in 56-day group.There were no statistical significances in all kinds of organ indexes among all groups in seven-day group.(ⅱ) There were no statistical significances in Hb among all groups. There were statistical significances in T between high=dose group and negative control group in seven-day group and it returned to be normal after one and two weeks of recovery.There were no statistical significances in FSH,LH and T among all groups in 56-day group.After 56 days of intragastric administration,there were statistical significances in ALT and AST between the high-dosegroup and other groups,in ALP between the low-dose group and negative control group,in ALB between the high-dose group and negative control group or high-dose group and medium-dose group,in A/G between the medium-dose group and negative control group(P<0.05).After one week of recovery,there were statistical significances in ALB and GLO between the high,dose group and low-dose group.After 2 weeks,there were no statistical significances in all serum biochemicals among all groups.(ⅲ) There were no statistical significances in the activity of SOD,GSH-Px,MDA and T-AOC in seven-day group.In 56-day group,after intragastric administration,there were no statistical significances in SOD,GSH-Px,MDA and T-AOC among all groups.After one week of recovery,there were statistical significance in GSH-Px between the high-dose group and other groups.After two weeks of recovery,there were statistical significance in GSH-Px between the high-dose group and control group,T-AOC between high-dose group and low-dose,control group.(ⅳ) After intragastric administration,significant pathological alterations were not studied with lightmicroscope.The changes of ultramicrostructure were observed with electronmicroscope and the main manifestations were that granulations increased and chondriosome dilated in Leydig cells,chondriosome and endocytoplasmic reticulum dilated in sertoli cells and lots of vacuolizations in spermatids.The injury in 56-day group was more serious than that in 7-day group.After recovery,The injury of 7-day group was more serious,56-day group slightly recovered.(ⅴ) After 56 days of intragastric administration,there were statistical significances in the percentage of 2C between the high-dose group and medium-dose group,4C between the high-dose group and control group,seminiferous epithelium cells at the cell-circle phase of S between the high-dose group and low-dose group,medium-dose group,seminiferous epithelium cells at the cell-circle phase of G2/M between the high-dose group and other groups,the 4C:2C ratio between the high-dose group and control,medium-dose group.After recovery,there were no statistical significances among all groups.Conclusion(ⅰ) Celery has some effect on liver function in male mice,but it can recover to be normal.(ⅱ) Celery has no effect on Hb,decreases the content of T in serum and change blood biochemical indexes.(ⅲ) Celery can increase the activity of antioxidation.(ⅳ) Celery has some effect on ultramicrostructure of testis in male mice,but it is reversible after two weeks of recovery.(Ⅴ) Celery can inhibit the spermatogenesis in mice.
