The Study On The Effect Of Reproductive System Induced By Celery In Famale Mice

Abstract/Summary:
Objective To explore the influences of celery on the reproductive system of female mice and its possible mechanisms,offering clues for the further development of celery.Methods The vaginal smears(once a time for each mice)of the 97 newly bought SPF strain adult female mice were checked at 8:00 every morning continuously for 14 days.Then the 97 mice were randomly separated into control group(only given NS), low-dose group,middle-dose group and high-dose group.Control group and other three different dose groups were given 0.3ml NS and 0.3ml different concentrations of celery respectively juice through intragastric administration each day.The changes of their estrous cycles were observed in the following days,and the mice were killed and the blood was collected on the 7th day and 14th day since intragastric administration. The hemoglobin was detected by Hb201+ HEMOCUE SWEDEN,the level of estrogen(E2),follicle-stimulating hormone(FSH)and luteotrop(h)ic hormone(LH)in blood was detected by RIA(radioimmunoassay),the contents of MDA,SOD and GSH-Px blood were detected by spectrophotometry,the tissue structure of mice's ovaries were observed through light microscope and electron microscope,and the expression of the Bcl-2 and Bax protein were detected by.Results(1)There were no statistical significances of weight among the different dose groups.The ovaries organ coefficient of the high-dose group was significantly higher than the control group's after being given celery juice through intragastric administration(P=0.014).The content of hemoglobin in the blood of high-dose group was significantly lower than the control group's after intragastric administration for 7 days.(2)No statistical significances of the total days ofestrous cycles of mice were found among different dose groups,but the time of pro-oestrus of mice in high-dose group afterl4 days intragastric administration of celery juice was found to be shorter than before(P=0.002),while the time of metaoestrus was found to be prolonged (P=0.018).There were no statistical significances of E2,FSH and LH among the four groups.(3)The activity of SOD in high-dose group was significantly higher than the control group after intragastric administration of celery juice for 7 days(P=0.013). The activities of SOD and GSH-Px in high-dose group were significantly higher than the control group(P=0.047,P=0.014),while the activity of MDA was significantly lower than the control group(P=0.042),after intragastric administration of celery juice for 14 days.(4)There were no abnormal signs in the structure of ovaries under light microscope,while different degrees of edema were found in the cytochondriomes of EMC and GrC of ovaries after given celery juice through intragastric administration for 7 days and 14 days under electric microscope.After 7 days,there were bold signs of necrosis in the follicular GrC in high-dose group,while after 14 days intragastric administration,dissolution and demolition were found in many places on the striated membrane of secondary follicle.(5)There were statistic differences of the protein expression of Bcl-2 and Bax between high-dose group and control group after 7 days intragastric administration(P=0.019,P=0.032),while after 14 days intragastric administration of celery juice,there were statistic differences of the protein expression of Bcl-2 and Bax between high-dose group and control group and between middle-dose group and control group(P=0.001,P=0.003;P=0.004,P=0.017).Conclusions Celery had some influence on female mices' estrous cycle, organizational structure of ovaries ability of anti-oxidation,and it can induce granular cell apoptosis.
