Objective To investigate the expression of centrosome-related kinase Nek2 in gastric cancer,to analyze its effect on the invasion of gastric cancer,and to search a new target for clinical diagnosis and treatment for gastric cancer.Methods Cancer samples and surgical margin of stomach were collected from 50 patients who underwent surgery in the general hospital of Tianjin medical university. Real time quantitative PCR and immunohistochemistry were used to detect the expression of Nek2 in these samples.Then to analysis the express of Nek2 correlate with the patients' clinicopathological characteristics.Results The expression of Nek2 mRNA and protein in cancer were significantly higher than those in tissues of surgical margin of stomach(P<0.05).There was significantly correlation between the expression of Nek2 mRNA and protein in cancer of TNM stage,depth of primary tumor invasion,differentiation of tumor and lymph node metastasis(P<0.05),but had no correlation with age,sex and the position of tumor(P>0.05).The Nek2 mRNA amplification copies and lymph metastasis rates present a positive correlation(r=0.870,p=0.000).Conclusion The Nek2 is overexpressed in gastric cancer on both mRNA and protein level,which indicates that Nek2 could be involved in development of tumors. The mRNA and protein levels of Nek2 in tumor tissue were positively correlated with the differentiation of tumor,the depth of tumor,lymphatic node metastasis as well as TNM stage of gastric carcinoma.The correlation between lymph node metastasis and the expression of Nek2 in gastric cancer indicates that Nek2 facilitates tumor progression.The Nek2 expression was independent risk factor for prognosis, which might be used in prognosis evaluation and clinical treatment for patients with GC to improve survival.
