ObjectiveAdolescent idiopathic scoliosis(AIS) is a common disease affecting adolescent health worldwide,which is the fifth in adolescent,after myopia,obesity,phimosis and psychologic obstacle.The etiology of it is unknown up to the present,but the hotspot of the research is concentrated on neuroendocrine system,especially the melatonin and calmodulin.It has been proved that the reduction of melatonin is one of significant causes,and the calmodulin has positive correlation to the severity of scoliosis.Animal models are the foundation of etiology research.Many animal models have been successfully made according to the known causes,but the way to make an ideal model is not been found.In this study,we want to obtain a scoliosis model in a way of injecting the antagon of melatonin receptor to abdominal cavity in bipeal rats,which may provide a relation between the melatonin,calmodulin and adolescent idiopathic scoliosis,and establish the experimental foundation of researching the etiology further.MethodsThirty healthy and weaning SD rats,which are fed for a week,are divided to five groups randomly,six rats each group.The groups of A,B,C,D are made up of bipedal rats,whose anterior limbs and tails are truncated.The groups of A,B,C are given the antagon of melatonin receptor,luzindole(ip) from 6pm to 8pm every evening.The doses of luzindole are respectively 0.2mg/kg,0.5mg/kg and 1mg/kg,and the doses of normal saline are all lOml.The group of D is given the same volume normal saline(ip) at the same time.The group of E is made up of quadruped rats,as blank comparison.The venous blood of all groups is drawn before they are executed.The thrombocyte is dissociated by centrifugal separation,and the calmodulin from thrombocyte is mensurated by the method of ELISA.Results1.The groups of A,B,C have all developed scolisis,especially the group of A,and scoliosis is present in 38.9%.The groups of D,E have neither developed scolisis, scoliosis is unpresent in 100%. 2.The group of C,given the most dose of the antagon of melatonin receptor is the most heaviest in all groups.3.There are no significant differences in the calmodulino of all groups,which are from the thrombocyte(p>0.05).Conclusions1.In our study,the scoliosis models of bipedal rats were found successfully,which could be reproduced easily,and confirmed that the generation of scoliosis was concerned with the reduction of the melatonin.Using the 0.2mg/kg dose of the antagon of melatonin(luzindole) can create the scoliosis model successfully.2.Luzindole,the antagon of melatonin receptor could promote the growth of rats.3.The level of calmodulino from thrombocyte had no relation with adolescent idiopathic scoliosis(AIS).
