| Objectives:To observe the effects of Huanglian Wendan Decoction on laboratory MS mice and its mechanism,and thus provide foundation for Huanglian Wendan Decoction on treating MS with Traditional Chinese Medicine and Materia medica.Methods:We rendered mice MS by feeding high-lipid,high- glucose, high-salt diet and then treated it with Huanglian Wendan Decoction, comparing to pioglitazone.Then we observed the effects of the drugs on weight,glyco metabolism,lipid metabolism,insulin sensitivity and blood pressure,Serum IL-6 and VCAM-1 also should be test to definite the mechanism that Huanglian Wendan Decoction may perform in MS rats.Results:①We succeeded in rendering laboratory MS,manifested as overweight,hypertension,hyperglycemia,hyperinsulinemia,high level of TG and lower level of HDL-C.With drug interference,the laboratory results are improved correspondingly,proving the animal model valid.②Serum tests of glycometabolism showed that Huanglian Wendan Decoction can significantly lower fasting blood sugar and insulin and thus improve IR condition,similar to the effect of metformin.Serum tests of lipid metabolism showed that Huanglian Wendan Decoction can significantly lower the level of serum cholesterol,triglyceride and low-density lipoprotein and lower the level of blood pressure and the weight of MS mice.Their function in regulating lipid metabolism and in adjusting blood pressure are better of to that of metformin.③Huanglian Wendan Decoction can significantly reduce serum IL-6 level(P<0.01).Immunohistochemistry studies of the effect of the drug on VCAM-1 in MS mice indicates that the drug can significantly lower the expression of VCAM-1 on the aorta in MS mice(P<0.01),comparable to metformin.Conclusions:1.It can build MS model with feeding high-fat high-sugar,high-salt diet to the rats. 2.Huanglian Wendan Decoction can loss weight,lower blood pressure, regulate lipid and improve insulin resistance,which can improve the various components of MS.3.Huanglian Wendan Decoction may improve MS by protecting the function of endothelial with its effect of reducing the serum IL-6 and lowering the expression of VCAM-1. |
