Objective:In the present study,we observed the change of blood glucose and the expression of AMPK protein in the early phase of acute endotoxemia in rats and explored the relation of relation between change of blood glucose and AMPK in the early phase of acute endotoxemia in rats.Methods:The rat model of endotoxemia was established with injecting lipopolysaccharide(LPS)through caudal vein.Healthy eight-week-old wistar rats(n=12) were randomly divided into control group and endotoxemia group.Blood glucose was determined at 0h,0.5h,1.0h,1.5h and 2h.Blood insulin was determinded at 2h.The expression of AMPK protein in myocardium,liver,soleus and extensor digitorum longus in the two groups was detected by Western blot analysis.Results:(1)After the LPS injection,the blood glucose level was significant higher than that of control group(P<0.05 or P<0.01).The increase appeared at 0.5h,reached the peak at 1.5h after the LPS injection,and then gradually decreased.(2) There was no significant difference between endotoxemia group and control group in the blood insulin level at 2h after the LPS injection(P>0.05).(3)The expression of Phos-AMPK a protein and AMPK a protein were no significant difference in myocardium between endotoxemia group and control group at 2h after the LPS injection(P>0.05).(4)The expression of Phos-AMPK a protein and AMPKαprotein were no significant difference in liver between endotoxemia group and control group at 2h after the LPS injection(P>0.05).(5)The expression of Phos-AMPKαprotein in soleus was significant higher than that of control group at 2h after the LPS injection (P<0.01).The expression of AMPKαprotein was no significant difference between them(P>0.05).(6)The expression of Phos-AMPKαprotein in extensor digitorum longus was significant higher than that of control group at 2h after the LPS injection (P<0.01).The expression of AMPKαprotein was no significant difference between them(P>0.05).Conclusions:(1)The blood glucose level increased at first and then gradually decreased in the early phase of acute endotoxemia in rats.(2)The decrease in the blood glucose level was no related to the blood insulin level in the early phase of acute endotoxemia in rats.(3)The expression of AMPKαprotein in myocardium,liver, soleus and extensor digitorum longus was not inhibited in the early phase of acute endotoxemia in rats.(4)The change in the blood glucose level was no related to myocardial or hepatic glycometabolism in the early phase of acute endotoxemia in rats. (5)The change in the blood glucose level was related to skeletal muscle glycometabolism in the early phase of acute endotoxemia in rats by increasing activity of AMPK,which was no related to the skeletal muscle fiber type.
