Partâ… The effects of the IL-1Ra on the apoptosis of the intervertebral disc cell of the bipedal ratObjective:To explore the effects of the interleukin?1 receptor antagonist protein (rhIL-1Ra)on the cell apoptosis of the bipedal rats' intervertebral disc.Methods:To establish a degenerative model a total of 54 bipedal rats.The 54 rats were randomly divided into three groups:experimental group,blank-control group and negative-control group,each group has 18 rats.The experimental group rats were intraperitoneal injected with IL-1Ra(50 ng per kilogram of body weight) from the day of operation and repeated it on alternate days until they were killed,and the negative-control in which 0.9%NS of the same dosage as IL-1Ra were injected. The blank-control group without treatment.Six rats were killed from every group at one,three,six months post-operation respectively.The tissue of L4/5 discs were fixed and sliced.The apoptosis cells of the intervertebral disc were assayed with terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling(TUNEL) and the ultrastructure were examined by transmission electron microscopy,and the tissue of the L5/6 discs were assayed with flowcytometry.Results:The statistic analysis of three groups had no significant difference at a month post-operation(P >0.05). The apoptosis cells of the experimental group had obviously decreased than blank-control group and negative-control group at three months post-operation, the statistic analysis had significant difference(P <0.05). The statistic analysis of three groups had very significant difference at six months post-operation(P <0.01)。There is no significant different between blank-control group and negative-con trol group at 3,6month post-operation(P >0.05).Conclusions:IL-1Ra protein can inhibit the apoptosis of the intervertebral disc cell of the bipedal rat.
