Objective: Atrophic gastritis is common and frequently-occurring disease of digestive system. For the past few years, many people carry out more researches and investigations to this illness, make a great contribution on diagnosis and therapy. But there is scarce precisely curative effect or little side effect western medicine. Traditional Chinese medicine to treat this disease have distinct dominantposition. Jiedu Huoxue Prescription is composed of herba,coptidis rhizome,corydalis,angelica,danshen root et al. It has the effects to detoxify, promote blood flow, and regulate qi to alleviate pain. It is the empirical formula, which has good therapeutic efficacy on this disease. This research applies four-factor method composed of MNNG, sodium salicylate, hot saline, and irregular diet to duplicate the model of atrophic gastritis rats by intragastric administration. And give weimeisu sanjiuweitai and Jiedu Huoxue Prescription to prevent beforehand or treatment this disease. The aim is to verify the curative effect of Jiedu Huoxue Prescription by observing the changes of pathological morphology of gastric mucosa, endothelin-1 thromboxan B2 and prostaglandin F1αof blood plasm of atrophic gastritis rats. And then discuss the functional mechanism of Jieduhuoxue fang curing AG.Methods:70 healthy wistar rats,half male and half female, were randomly divided into 5 groups, 14 rats every group, named normal group, model group, Jiedu Huoxue Prescription group, sanjiuweitai group, weimeisu group. The normal group didn't to make model, give grounding feedstuff and distilled water (2ml/only/day) through intragastric administration everyone everyday for comparison; the other group freedom drinking 160μg/ml MNNG, 2% sodium salicylates and 15% NaCl solution freely (heating to 55℃) fills the stomach full alternately every other day, irregular diet (Overeating for two days, starvation one day), to made atrophic gastritis rats model. The medication:take MNNG dissolve in distilled water, to mate solution of density 4.17g/L, away from ligh 4 oC cold-storage to maintain. Before to use, dilute it to 160μg/ml by distilled water, and insertion to black watter bottle freedom drinking (270ml/only/day), to change everyday for 12 weeks. Jiedu Huoxue Prescription group: to weighe the traditional Chinese medicine by prescriptiongives proportion, decoction and to condense two orders, according to 20 times of mankind dose to compute, give intragastric administration by the 51.7g/kg suspension (2ml/only/day) for 12 weeks. Sanjiuweitai group: making suspl by dissolving sanjiuweitai in distilled water according to 15 times of mankind dose to compute and gives intragastric administration by the 10ml/kg suspension (2ml/ only/day) for 12 weeks. Weimeisu group: making suspl by dissolving weimeisu in distilled water according to 15 times of mankind dose to compute and gives intragastric administration by the 0.99g/kg suspension (2ml/only/day) for 12 weeks. Before and after intragastric administration to absolute diet 2 hours. To observe general state of rats everyday. After 12 weekends, we draw each 2 (female and male each 1) from normal group and model group at random, and then we kill them, open the abdomen, take the stomach, especially we take each 1 tissue from sinus ventriculi and corpus ventriculi. Later we makes the light microscope observation fixedly with 10% formalin. Once the light microscope observation discovered the atrophy in big mouse's glandular organ of gastric mucosa, we finished making the mold. After the last time administration, we stopped giving the rats food and drink for 24 (gives water) hours. We cut the belly open to obtain stomach by aseptic technique, then sheared the stomach along arcus major ventriculi, rinsed gastric contents with ice-physiological saline, blotted it with filter paper and macroscopic observed it. At last, two pieces of tissues of sinus ventriculi were scissorsed, each piece about 0.5 cm3. The tissues were fixed with 4 percent paraform, dehydrolysised with alcohol, paraffin imbedding, and cut into sheets. And the sheets were dyed to observe pathological changes of gastric mucosa with light microscope.Take suction indomethacin EDTA.Na2 about 0.2 ml by a empty syringe of 5 ml, quickly venipuncture to obtained blood about 3ml and instantly put upside down the syringe to misce bene the liquid. Then put it in vitro, Centrifuged the blood (4oC3500rpm 15 minutes) and separated plasma to examine the content of thromboxan B2 and prostaglandin F1αlater. Examine thromboxan B2 use equilibrium radio-immunity and examine prostaglandin F1αuse nonequilibrium. Put them to death by decapitation and obtained blood 2ml. Added 7.5% EDTA-2Na 30μl and aprotinin 40μl to the blood and jolted in plastic tube.Centrifuged the blood (4oC3000rpm 10 minutes) and separated plasma to examine the content of endothelin-1 later. Examine endothelin-1 use nonequilibrium.Results:1. The effect of Jiedu Huoxue Prescription on general state of health of atrophic gastritis rats:General state of health: the mental state of normal group rats is very good, the fur is glossand and well-arranged; the mental state of model group is very poor, the food-intake obviously grow downwards the fur is confused and disorderly and dullness;the ingestion and haris of other three group of rats is improve follow the dosage. Especially, the ingestion of Jiedu Huoxue gpoup of rats is obviously grew than the other three group except the normal group. During the experiment, 2 rats were dead in each group: Jiedu Huoxue Prescription group, sanjiuweitai group ,weimeisu group.The reasons may be intragastric administration and drug purification and so on. 2. The effect of Jiedu Huoxue Prescription on pathological morphology of gastric mucosa of atrophic gastritis rats:Gross observation, normal control group Gastric mucosa were pink, shiny, covered with more mucus. And the surface of the mucosal folds were smooth and flexible which taken the rule shape. Model group Pale gastric mucosa thinned. Furthermore, the submucosal vessel could be seen easily. The mucosal folds were level or taken the disorder shape. And the elasticity of stomach wall decreased Sanjiu weitai and weimeisu control group The gastric mucosa thinned, to a greater or lesser degree, which were pale. And the elasticity of stomach wall decreased, to different extent. But the representation were better than other control group. Jiedu Huoxue Prescription group showed no significant difference between the normal control group, which was significantly superior to other control groups, to the naked eye observation.Optical microscope, normal control group Mucosal epidermis of rats was complete and the glandular organs array regularly. Lamina muscularis mucosae were thin. Model group mucosal epidermis of rats was complete and thin, of which the glandular organs array irregularly. Especially the glandular organs shrink back or disappear to different degree. And the mucous membrane tunica muscularis layer thickness were thickened with the mucous membrane extending to tunica propria. Gastric mucosa Jiedu Huoxue Prescription, the mucosal epidermis was complete. The layer thickness of the gastric mucosa were even. And the glandular organs array regularly. Sanjiu weitai control group mucosal muscularis were relatively thin. Not very few glandular organs shrink back. Weimeisu control group mucosal muscularis layer thickness were obviously thickened. Many glandular organs shrink back.3. The comparison of every group on thromboxan B2 and prostaglandin F1αof blood plasmCompared with normal group, the content of thromboxan B2 in model group was heigher, which had statistical significance (P<0.05); Compared with model group, the contents of thromboxan B2 in Jiedu Huoxue Prescription group, sanjiu group and weimeisu group were lower, which had statistical significance (P<0.05); Compared with the contents of thromboxan B2 in sanjiu group and weimeisu group, the Jiedu Huoxue Prescription group is obviously lower than the other groups, which had statistical significance (P<0.05).Compared with normal group, the content of prostaglandin F1αin model group was lower, which had statistical significance (P<0.05); Compared with model group, the contents of prostaglandin F1αin Jiedu Huoxue Prescription group, sanjiu group and weimeisu group were higher, which had statistical significance (P<0.05); Compared with the contents of prostaglandin F1αin sanjiu group and weimeisu group, Jiedu Huoxue Prescription group is obviously higher than the other groups, which had statistical significance (P<0.05).4. The comparison of every group on endothelin-1 of blood plasmCompared with normal group, the content of endothelin-1 in model group was higher, which had statistical significance (P<0.05); Compared with model group, the contents of endothelin-1 in Jiedu Huoxue Prescription group, sanjiu group and weimeisu group were lower, which had statistical significance (P<0.05); Compared with the contents of endothelin-1 in sanjiu group and weimeisu group, Jiedu Huoxue Prescription group is lower, which had statistical significance (P<0.05).Conclusion:1. Jiedu Huoxue Prescription has a certain curative effect. It could improve the changes of pathomorphology in gastric mucosa, relieve the damnification of gastric mucosa, to improve the atrophia and arrangement of gastric mucous membrane glandular organ. It has the function of protecting gastric mucosa of atrophic gastritis rats effectively.2. Jiedu Huoxue Prescription could degrade the content of thromboxan B2 in blood plasma, and increase the content of prostaglandin F1αin blood plasma, then raise the volume of blood flow of gastric mucosa, to improve the state of gastric mucosa. So it could be to lessen the damage of gastric mucosa.3. Jiedu Huoxue Prescription could degrade the content of endothelin-1 in blood plasma, then to improve the volume of blood flow of gastric mucosa, to improve ischemia of gastric mucosa, in favour of the absorption of infl and the restoration of glandular organ, then lessen the damage of gastric mucosa to protect it.
