Objective:Fucoidan is a kind of water-solubility heteroglycan that lies in many kinds of brown algaes and some oceanic invertebrates.In recent years,many studys prove that Fucoidan possesses excellent biological properties,such as anticoagulting,increasing immunity,lowering blood-fat and so on.But there are few reports on The effect of Fucoidan on the adhesion and aggregation of platelet.The aim to study the effect of Fucoidan on the adhesion and aggregation of platelet is to provide a new mentality on the anti-platelet medicine research and a valuable scientific basis for the produce of high-valued functional food and medicine from Laminaria.The social and economic value is substantial.Methods:Adult New Zealand rabbit were randomly divided into four groups:the blank control group(A group),the aspirin group(B group),the low dose of Fucoidan group(C group) and the high dose of Fucoidan group(D group).Rabbits were separately treated with normal food,Aspirin 10mg/kg,Fucoidan 10mg/kg and Fucoidan 100mg/kg at one week before drawing blood).Then the platelet adherence test(PadT) and the platelet aggregation test(PagT) were separately examined so that the adherence ratio,the most aggregation ratio of the platelet and the level of the Thromboxane B2were achieved.Results:The 7th day after taking medicine,there was not significant difference on the adherence ratio of the platelet between the B group (30.99±8.73%) and the blank control group(A group,34.85±10.17%) (q=1.345,P>0.05).The adherence ratio of the platelet of the C group and D group(16.08±6.26,16.74±6.72) were lower than the blank control group(A group,34.85±10.17).The difference was significant(q=18.7750,q=18.1125, P<0.05).The most aggregation ratio of the platelet of the B group(49.81±8.87) was lower than the blank control group(A group,71.86±10.88).The difference was significant(q=7.023,P<0.05).The most aggregation ratio of the platelet of the C group and D group(61.78±7.98,58.81±7.41) were lower than the blank control group,but higher than the B group.The difference was significant(q=3.808,q=2.965,P<0.05).The level of the Thromboxane B2of the B group(14.06±3.54ng/L) was lower than the blank control group (A组,137.18±33.77ng/L).The difference was significant(q=12.67,P<0.05).There was not significant difference on the level of the Thromboxane B2 between the C group and D group(130.13±30.54ng/L, 135.14±30.56ng/L) and the blank control group(A group,137.18±33.77ng/ L)(q=0.73,q=0.21,P>0.05).There were not significant difference on the adherence ratio,the most aggregation ratio of the platelet and the level of the Thromboxane B2 between all the groups before taking medicine and on the 7th day after taking medicine.Conclusions:The inhibition of Fucoidan on the adherence of the platelet is significant.The is also a certain degree of inhibition on the platelet aggregation.However,the effect of Fucoidan is weak than aspirin. There is not relational between the inhibition of platelet aggregation and the level of thromboxane B2.
