Aims:To study the effect and mechanism of acute exercise on IGF-ImRNA and its protein synthesis in rat skeletal muscle,it tested the effect of acute exercise on rat IGF-1 and its protein expression in different intensions and durations.Materials and methods:SD rats were randomly divided into four groups:normoxia control group,low,moderate,high intensity,the moderate group was divided into three groups:30,60,9Omin,the exercise groups were divided and killed in three times after acute exersise in lh,6h,12h.IGF-ImRNA and its protein expression in quadriceps muscle were tested with the method of RT-PCR,ELISA.Results:(1)after different intensive acute exercise IGF-ImRNA and its protein expression increased significantly(p<0.05),and its expression increases with the intensive exercise(p<0.05);(2) IGF-lin the same group,there are no differences in 1,6,12;(3)After moderate exercise in different duration.IGF-ImRNA and its protein expression increased significantly(p<0.05).No difference in IGF-1 of the three groups which were killed in the same time.No difference in IGF-1 of the groups in the same intension which were killed 1,6,12 after the exercise. (4) IGF-1 mRNA have no difference among the groups killed in the same time(p>0.05).In the duration groups,IGF-1 mRNA in the group which was killed 6h after exercise is significant higher than the groups killed 1h,12h.(p<0.01)Conclusions:(1)different intensive acute exercise,promote the synthesis of IGF-1 and IGF-1 mRNA,and increase with the intension.(2) the synthesis of IGF-1 increased significantly and keep the high level from 1 to 12 after exercise.The synthesis of IGF-1 mRNA keep increasing to the highest level in 6h after exercise,then began to fall until 12h after exercise;(3)Moderate exercise in different duration promot the synthesis of IGF-1 mRNA and IGF-1,the duration have no significant impact on them.
